Emerging evidence suggests that E3ligases play critical roles in diverse biological processes, including innate immune responses in plants. However, the mechanism of the E3ligase involvement in plant innate immunity is unclear. We report that a rice gene, OsBBI1, encoding a RING finger protein with E3ligase activity, mediates broad-spectrum disease resistance. The expression of OsBBI1 was induced by rice blast fungus Magnaporthe oryzae, as well as chemical inducers, benzothiadiazole and salicylic acid. Biochemical analysis revealed that OsBBI1 protein possesses E3 ubiquitin ligase activity in vitro. Genetic analysis revealed that the loss of OsBBI1 function in a Tos17-insertion line increased susceptibility, while the overexpression of ...

Abstract The heterotetrameric K++-channel KCNQ1/KCNE1 is expressed in heart, skeletal muscle, liver and several epithelia including the renal proximal tubule. In the heart, it contributes to the repolarization of cardiomyocytes. The repolarization is impaired in ischemia. Ischemia stimulates the AMP-activated protein kinase (AMPK), a serine/threonine kinase, sensing energy depletion and stimulating several cellular mechanisms to enhance energy production and to limit energy utilization. AMPK has previously been shown to downregulate the epithelial Na++ channel ENaC, an effect mediated by the ubiquitin ligase Nedd4-2. The present study explored whether AMPK regulates KCNQ1/KCNE1. To this end, cRNA encoding KCNQ1/KCNE1 was injected into Xenopus oocytes with and without additional injection o...
The inducible transcription factor NF-#kappa#B regulates divergent signaling pathways including inflammatory response and cancer development. Selective inhibitors for NF-#kappa#B signaling are potentially useful for treatment of inflammation and cancer. NF-#kappa#B is canonically activated by preferential disposal of its inhibitory protein; I#kappa#B, which suppresses the nuclear translocation of NF-#kappa#B. I#kappa#B#alpha# (a major member of I#kappa#B family proteins) is phosphorylated with an I#kappa#B kinase (IKK) and subsequently polyubiquitylated by SCF"#beta#"T"r"C"P"1 ubiquitin-ligase in the presence of E1 and E2 prior to proteasomal degradation. Here, we describe a novel inhibitor termed GS143, which suppressed I#kappa#B#alpha# ubiquitylation, but not I#kappa#B#alpha# phosphorylation, MDM2-directed p53 ubiquitylation, and proteasome activity in vitro. GS143 markedly suppressed the destruction of I#kappa#B#alpha# stimulated by TNF#alpha# and a set of ...

The E1 and E3 transition operators in the sdf-IBA model are discussed. An effective E1 operator containing one- and two-body terms explains very well the E1 data in transitional and rotational nuclei. The E1 parameters for rotational nuclei are obtained by imposing a constraint on the IBA model namely, the validity of Alaga rules for ``pure K`` states. The systematic behavior of the octupole strength is well reproduced using a one-body octupole operator. The anomalously large fragmentation of the low-energy octupole strength in non-rotational nuclei is shown to be a signature of the O(6) dynamical symmetry.
Abstract High-mobility group box 1 (HMGB1), a damage-associated molecular pattern (DAMP) molecules, favors tissue regeneration via recruitment and activation of leukocytes and stem cells. Here we demonstrate, in a model of acute sterile muscle injury, that regeneration is accompanied by active reactive oxygen species (ROS) production counterbalanced and overcome by the generation of antioxidant moieties. Mitochondria are initially responsible for ROS formation. However, they undergo rapid disruption with almost complete disappearance. Twenty-four hours after injury, we observed a strong induction of MURF1 and atrogin-1 ubiquitin ligases, key signals in activation of the proteasome system and induction of muscle atrophy. At later time points, ROS generation is maintained by nonmitochondrial sources. The antioxidant response occurs in both regenerating fibers and leukocytes that express high levels of free thiols and antioxidant enzymes, such as superoxide dismutase ...
We discuss an experimental technique aimed at tagging electron neutrinos in multi-GeV artificial sources on an event-by-event basis. It exploits in a novel manner calorimetric and tracking technologies developed in the framework of the LHC experiments and of rare kaon decay searches. The setup is based on an instrumented decay tunnel equipped with tagging units that intercept secondary and tertiary leptons from the bulk of undecayed pi+ and protons. We show that the taggers are able to reduce the nue contamination originating from K_e3 decays by about one order of magnitude. Only a limited suppression (~60%) is achieved for nue produced by the decay-in-flight of muons; for moderate beam powers, similar performance as for K_e3 can be reached supplementing the tagging system with an instrumented beam dump.

This paper describes an experiment which has been performed with a somewhat different philosophy in its approach as compared to previous work. (a) The trigger threshold energy was increased by more than a factor of ten on previous experiments reducing the dilution effect of many lower energy showers known not to produce tachyons. 1673 shower were recorded. (b) The exposure time was made longer than previous experiments in order to obtain a reasonably large statistical sample at the higher threshold. Observations were made between February 1979 and June 1980, the total live time being 9768 hours. (c) Records of the detector output amplitude for the 480#mu#s lead time were integrated point by point as is sometimes done in radio astronomy, this gives the integrated energy deposition within the detector.
In the field of Living Probabilistic Safety Assessment (LPSA) the reliability data updating is an important factor. In risk analysis equipment failure data is needed to estimate the frequencies of events contributing to risk posed by a facility. Five years data of emergency diesel generator (EDG) of Daya Bay Nuclear Power Plant (NPP) has been studied in this paper. The data updating process has been done by using two methods, i.e., the classical method and Bayesian method. The aim of using these methods is to calculate the operational failure rate (@l) and demand failure probability (p). The results show that the operational failure rate is 1.7E-3 per hour and the demand failure probability is 2.4E-2 demand per day for Daya Bay NPP. By comparing the results obtain from classical and Bayesi...
Gamma ray spectra in the decay of 185Ta and 185mW have been studied with Ge (Li) detectors. The 185mW isomeric transition at 131.6 keV is shown to be of E3 multipolarity. A level scheme of 185W is proposed with the following energy levels (energies in keV, spin and K quantum numbers in brackets): 0 (3/2- 3/2), 23.5 (1/2- 1/2), 65.9 (5/2- 3/2), 93.5 (3/2- 1/2) (uncertain), 173.9 (7/2- 3/2), 188.1 (5/2- 1/2), 197.5 (11/2+ 11/2) , 243.5 (7/2- 7/2), and 390.8 (9/2- 7/2)
Net heats of reaction were measured in an isothermal calorimeter for both single phase (organic) and two phase (organic and aqueous) TBP/HNO{sub 3} reacting solutions at temperatures above 100 C. The oxidation rate constant was determined to be 5.4E-4 min{sup {minus}1} at 110 C for an open ``vented`` system as compared to 1.33 E-3 min{sup {minus}1} in the closed system. The heat released per unit material oxidized was also reduced. The oxidation in both phases was found to be first order in nitric acid and pseudo-zero order in butylnitrate and water. The hydrolysis (esterification) rate constant determined by Nichols` (1.33E-3 min{sup {minus}1}) fit the experimental data from this work well. Forced evaporation of the volatile components by the product gases from oxidation resulted in a cooling mechanism which more than balanced the heat from the oxidation reaction in the two-phased systems. In the report the results of the estimations of radiological risk of various stages of decommissioning of nuclear submarines are presented. At occurrence on nuclear submarine the heavy failure, relating to the class hypotetical volume of acting of radionuclides in atmosphere can reach 1.6E(15) Bq. Results of estimations probable doses on an axis of a trace of a radioactive loop show, that at distribution of radionuclides during atmospheric carry to 'agreed' settlement (500-1000 m) the maximum doses on its territory can make: about 6.0E(-3) Sv (for the whole body); 3.0E(-3) Gy for the leather (basal layer); 6.3E(-2) Gy for the lungs (acute exposure) and up to 1.8 Gy for the thyroid gland. Hypotetical failure for the estimation of the greatest possible radioecological consequences for hydrobiocenosis is considering, connected with single discharge of liquid radioactive waste (LRW) in water area. TIGER integral-field spectrography and HST/WFPC2 imaging of the E3 galaxy NGC 2974 are used to derive the kinematics of the stellar and ionized gas components in its central 500 pc. We derive a numerical two-integral distribution function from a MGE mass model using the HQ formalism. The TIGER as well as published long-slit stellar kinematics are well fitted with this self-consistent model, requiring neither the addition of a significant mass contribution from a hidden disc structure, nor the presence of a central dark mass. The data reveal the presence of a striking, highly contrasted, two-arm gaseous spiral structure within a radius of ~200 pc, corresponding to a total mass of 6.8x10^4 Msun of ionized gas. We use a deconvolved TIGER datacube to probe its kinematics at a resolution of about 0.35 arcsec. Strong departures from circular motions are observed, as well as high velocity dispersion values on the inner side of the arms. We interpret ...
Purpose: The aim of this study was to test the feasibility of iliac artery stent placement under MR guidance with real-time MR radial scanning in an animal model. Materials and methods: The experiments were performed on three pigs in a 1.5 T scanner. Radial scanning with a gradient echo technique (T{sub R} 8.4 ms, T{sub E} 3.6 ms, flip angle 10 ) was used. A dedicated backprojector performed the reconstruction of the raw data in real-time. The resulting MR-images were displayed on LCD screens beside the magnet. The sliding window reconstruction technique allowed image acquistion at a frame rate of 16 images per second. MR-compatible self-expanding stents with a diameter of 8 mm and a length of 3 cm were placed into the left iliac artery. Their positions were verified by digital subtraction angiography (DSA) and compared to MRI. Results: All stents were successfully placed. Stent positions as monitored by real-time MR were identical to those ...
The purpose of this study was to examine arachidonic acid (AA) metabolism and to characterize the type of adrenergic receptor (AR) involved in the production of the major metabolite of this fatty acid. ({sup 14}C)AA was incubated with GPT-rings and the radiolabelled products were extracted and separated by TLC method. The medium was also assayed for radiolabelled immunoreactive PG's (iPG's) and leukotrienes (LT) B4 and C4 by RIA or Enzyme immunoassay (EIA) after exposure to various AR agonists. ({sup 14}C)AA was incorporated into GPT-rings and metabolized mainly into iPGE2 and smaller amounts into PGF2{alpha}. Trace amounts of PGD2 and 6-keto-PGF1{alpha} but not LTB4 or LTC4 were detected by RIA and/or EIA. Incubation of GPT rings for 15 minutes with isoproterenol and salbutamol resulted in a significant increase of PGE2 synthesis (optimum conc: 10{sup {minus}7}, 10{sup {minus}7}M respectively). Thermoluminescence (TL) and Electron Paramagnetic Resonance (EPR) dosimetry were used to measure dose effects in borosilicate glass with time, from 10 minutes to #approx#60 days following exposure to a dose of up to 10,000 Rad. TL and EPR results were consistent and performed similarly, with both techniques capable of achieving an estimated limit of detection of between 50-100 Rad. Three peaks were identified in the TL glow curve at roughly 110 C, 205 C, and 225 C. The intensity of the 205 C peak was the dominant peak over the time period of this study. The stability of all of the peaks with time since irradiation increased with their corresponding temperature and little or no variation was observed in the glow curve response to a specified total dose attained at different dose rates. The intensity of the 205 C peak decreased logarithmically with time regardless of total dose. Based upon a conservative limit of detection of 330 Rad, a 10,000 Rad dose would still be detected ...
The waste tail gas fuel emitted from refinery plant in Taiwan e.g. catalytic reforming unit, catalytic cracking unit and residue desulfurization unit, was recovered and reused as a replacement fuel. In this study, it was slowly added to the fuel stream of a heater furnace to replace natural gas for powering a full-scale distillation process. The waste tail gas fuel contained on average 60 mol% of hydrogen. On-site experimental results show that both the flame length and orange-yellowish brightness decrease with increasing proportion of waste gas fuel in the original natural gas fuel. Moreover, the adiabatic flame temperature increases as the content of waste gas fuel is increased in the fuel mixture since waste gas fuel has a higher adiabatic flame temperature than that of natural gas. The complete replacement of natural gas by waste gas fuel for a heater furnace operating at 70% loading (i.e. 3.6 x 10{sup 7} kcal/h of combustion capacity) will ...
The analgesic activity of nine substituted N-pyrrolylcarboxylic acids, previously reported as anti-inflammatory agents, has been evaluated. The effects on nociception were examined in male Wistar rats by the Randall-Selitto paw-pressure test. The compounds were administrated in doses 10, 20, and 40 mg/kg both i.p. and p.o. As a whole, the activities of 3-(N-pyrrolyl)propanoic acids 3e-3h and 2-[3-(ethoxycarbonyl)-2-methyl-5-phenyl-1H-1-pyrrolyl]-3-methylpentanoic acid 3i were comparable with or superior to that of metamizole used as a reference (200 mg/kg, i.p.), whereas only 3a from among the N-pyrrolyl-acetic acids 3a-3d showed analgesic activity on the inflamed paw. The compounds found most promising to increase the pain threshold significantly were the same ones with the higher anti-inflammatory activity registered in our previous study: 3-[3-(ethoxycarbonyl)-2-methyl-5-(3-nitrophenyl)-1H-1-pyrrolyl]propanoic acid 3e, together with its ...
Up to the present, the generic values of the Common cause failure (CCF) event parameters have been used in most PSA projects for the Korean NPPs. However, the CCF analysis should be performed with plant specific information to meet Category II of the ASME PRA Standard. Therefore, we estimated the Alpha factor parameters of the emergency diesel generator (EDG) for Ulchin Unit 3 by using the International Common-Cause Failure data Exchange (ICDE) database. The ICDE database provides the member countries with only the information needed for an estimation of the CCF parameters. The Ulchin Unit A3, pressurized water reactor, has two onsite EDGs and one alternate AC (AAC) diesel generator. The onsite EDGs of Unit 3 and 4 and the AAC are manufactured by the same company, but they are designed differently.
