TGN had a rupture in service on Gasoducto Troncal Norte. After initial evaluation of the causes of the rupture it was concluded that it had been caused by Stress Corrosion Cracking (SCC). Subsequent investigation in the area of the rupture revealed that colonies of cracks, typical of SCC were found in pipes located near the rupture. In order to put back in service the pipeline in a safety condition, SCC mitigation activities were performed. A decision was made to conduct a hydro test along approximately 30 kilometers of pipe. The stages of the works, the problems faced and the solutions found are dealt with, as well as the conclusions reached upon completion of the works which allowed a better understanding of SCC phenomenon. The methodology for the identification of those areas susceptible to SCC is also described. (author)

So far, it is believed that they degenerate with progression of HIV disease because deterioration of DA systems is evident in advanced stages of infection. In this manuscript we found that (a) DA levels are increased and DA turnover is decreased in CSF of therapy-nave HIV patients in asymptomatic infection, (b) DA increase does not modulate the availability of DA transporters and D2-receptors, (c) DA correlates inversely with CD4+ numbers in blood. Micro-organisms that degrade plant biomass require a range of enzymes to effectively degrade such substrates into sugar monomers. Some organisms secrete a host of free, extracellular enzymes while others produce a multi-enzyme complex (MEC) consisting of a variety of these enzymes. This study examined the cellulolytic and hemi-cellulolytic system of Bacillus licheniformis SVD1 with respect to the presence of key enzymes within the MEC found in this organism. When cultured on birchwood xylan, the MEC in B. licheniformis SVD1 was found to contain two endoglucanases (at 25-27kDa, 30-32kDa), seven xylanases (at 20kDa, 38kDa, 42kDa, 45kDa, 48kDa, 54kDa and 70kDa), two mannanases (25kDa, 40-42kDa) and one pectinase (a pectate lyase at 70-72kDa). Dopamine (DA), apomorphine and B-HT 933 produced dose related contractions on isolated longitudinal strips of chicken esophagus, whereas phenylephrine elicited no effect. DA induced contractions of myogenic origin, these contractions were insensitive to DA antagonists and were partially suppressed by yohimbine, which suggested an alpha2-adrenergic implication in this DA effect. This hypothesis was further investigated by performing binding experiments, in which B-HT 933 displaced the binding of [3H] DA to esophageal homogenates. In the present article, we have investigated the dopamine (DA) content, homovanillic acid (HVA)/DA turnover ratio, DA D1 and DA D2 receptors in the hippocampus of insulin-induced hypoglycaemic (IIH) and streptozotocin induced diabetic rats where brain functions are impaired. The expression `dental age' (DA) denotes the mean age at which a given stage of dental development is achieved in a population chosen as calibrator. The rhythm at which the maturation occurs is a characteristic of a given population in a given period. When DA is determined on subjects of a population different from the calibrator, the shape of the relationship of DA to chronological age (CA) must be empirically determined to trace reference charts for DA that properly apply to this population. PanDA is a pilot-based production and distributed analysis system developed and used extensively by ATLAS. This work presents the implementation and usage experiences of a PanDA backend for Ganga. Built upon reusable application libraries from GangaAtlas and PanDA, the Ganga PanDA backend allows users to run their analyses on the worldwide PanDA resources, while providing the ability for users to develop simple or complex analysis workflows in Ganga. Further, the backend allows users to submit and manage "personal" PanDA pilots: these pilots run under the user's grid certificate and provide a secure alternative to shared pilot certificates while enabling the usage of local resource allocations

Isolation and cDNA cloning of ovarian cortical rod protein in kuruma prawn Marsupenaeus japonicus (Crustacea: Decapoda: Penaeidae).

Two cortical rod proteins having molecular weights of 28.6 kDa and 30.5 kDa were isolated from the mature ovary of Marsupenaeus japonicus using gel filtration and reversed-phase HPLC. Analysis of the N-terminal amino acid sequence of the 28.6 kDa molecule revealed that amino acid residues 1-21 corresponded to residues 9-29 of the 30.5 kDa molecule. Examination of homology using BLAST showed that 21 amino acids out of 29 residues of the 28.6 kDa molecule, and 14 out of 29 residues of the 30.5 kDa molecule were identical to that of the ovarian cortical rod proteins of Penaeus semisulcatus. Positive immunohistochemical reaction to antiserum raised against the 28.6 kDa protein was observed on cortical rods forming around the periphery of oocytes at the maturation stages. Western blotting analysis revealed that both the 28.6 kDa and 30.5 kDa molecules stained with the anti-28.6 kDa antiserum. Furthermore, the 28.6 kDa and 30.5 kDa proteins were both glycosylated, as evidenced by positive carbohydrate staining using Concanavalin A and production of positive PAS reaction. These results indicate that the cortical rods are comprised of the 28.6 kDa and 30.5 kDa molecules. We subsequently cloned two full-length cDNAs based on the N-terminal sequences of the 28.6 kDa and 30.5 kDa molecules. The open reading frame of 28.6 kDa and 30.5 kDa encoded 276 amino acid residues. Comparison analysis of the two cDNAs revealed that the location of the processing site and sequence of signal peptides differed, indicating that the two cDNAs are products of two separate genes and encode the 28.6 kDa molecule and 30.5 kDa molecule, respectively. Both proteins possessed one potential N-linked glycosylation site. It is considered that both molecules are components of the cortical rods, forming a jelly layer after fertilization.

PI3K Signaling Supports Amphetamine-induced Dopamine Efflux

Full Text Available.The dopamine (DA) transporter (DAT) is a major molecular target of the psychostimulant amphetamine (AMPH). AMPH, as a result of its ability to reverse DAT-mediated inward transport of DA, induces DA efflux thereby increasing extracellular DA levels. This increase is thought to underlie the behavioral effects of AMPH. We have demonstrated previously that insulin, through phosphatidylinositol 3-kinase (PI3K) signaling, regulates DA clearance by fine-tuning DAT plasma membrane expression. PI3K signaling may represent a novel mechanism for regulating DA efflux evoked by AMPH, since only active DAT at the plasma membrane can efflux DA. Here, we show in both a heterologous expression system and DA neurons that inhibition of PI3K decreases DAT cell surface expression and, as a consequence, AMPH-induced DA efflux.

Spectroscopic analysis of DA white dwarfs from the McCook and Sion catalog

For some years now, we have been gathering optical spectra of DA white dwarfs in an effort to study and define the empirical ZZ Ceti instability strip. However, we have recently expanded this survey to include all the DA white dwarfs in the McCook and Sion catalog down to a limiting visual magnitude of V = 17.5. We present here a spectroscopic analysis of over 1000 DA white dwarfs from this ongoing survey. We have several specific areas of interest most notably the hot DAO white dwarfs, the ZZ Ceti instability strip, and the DA+dM binary systems. Furthermore, we present a comparison of the ensemble properties of our sample with those of other large surveys of DA white dwarfs, paying particular attention to the distribution of mass as a function of effective temperature

In Phase I we proposed the following tasks: Task 1: Identify the units of the Differential Algebra (DA) library, i.e. determine the abstract data types of the DA and the relations between them. Develop the interfaces (.h files) for the DA vectors. Task 2: Implement the DA vector class with garbage collection and expression templates for optimizing all overloaded operators by minimizing creation of temporaries and fusing loops. Task 3: Implement the prototype GUI for instantiating systems from files and invoking the DA methods. Task 4: Develop a suite of tests for the DA vector class and the needed utilities classes. Task 5: Write the final report on this work. This will include documentation on the use of the code. We have completed these tasks. In this section we discuss the results of our work.

We investigated the effect of ginseng total saponin (GTS) on nicotine-induced dopamine (DA) release in the striatum and nucleus accumbens of freely moving rats using in vivo microdialysis technique. Systemic pretreatment with GTS decreased striatal DA release induced by local infusion of nicotine into the striatum. However, GTS had no effect on the resting levels of extracellular DA in the striatum. GTS also blocked nicotine-induced DA release in the nucleus accumbens. The results of the present study suggest that GTS acts on the DA terminals to prevent DA release induced by nicotine. This may reflect the blocking effect of GTS on behavioral hyperactivity induced by psychostimulants.

An exploratory study for rapid estimation of critical source parameters of great subduction-zone earthquakes in Mexico

The rapid and reliable estimation of moment magnitude M{sub w}, location, and size of rupture area, and radiated energy E{sub s} of great Mexican subduction zone earthquakes is critical for a quick assessment of tsunami and/or damage potential of the event and for issuing an early tsunami alert. To accomplish this goal, the Mexican broadband seismic network needs to be supplemented by permanent GPS stations along the Pacific coast, spaced about 65 km apart or less. The data from the GPS network must be transmitted to a central location and processed in near-real time to track the position of the stations. Assuming that this can be implemented, we develop a procedure for near-real time estimation of the critical source parameters. We demonstrate the viability of the procedure by processing near-source GPS data and regional seismograms for the earthquakes of Colima-Jalisco in 1995 (M{sub w}=8.0) and Sumatra-Andaman in 2004 (M{sub w}=9.0-9.3). The procedure yields estimates of M{sub w} and E{sub s} in excellent agreement with those reported from earlier solutions. In the case of the Colima-Jalisco earthquake, the estimated location and size of rupture area agree with that mapped from aftershock locations. Presently, there are 13 permanent GPS stations along the Pacific coast of Mexico with an average spacing of {approx}200 km which operate in an autonomous mode. It is urgent to increase the number of stations to {>=}28 thus decreasing the spacing of stations to {<=}65 km. Data must be transmitted in near-real time to a central station to track the position of the stations, preferably every second. [Spanish] Para una estimacion oportuna del potencial de dano y tsunami asociado a los grandes temblores de subduccion en Mexico, resulta critica la determinacion rapida y confiable de parametros sismologicos como lo son la magnitud de momento (M{sub w}), la energia sismica radiada (E{sub s}) y la localizacion y el tamano de la ruptura. Para alcanzar este objetivo, la red sismologica de banda ancha mexicana necesita ser complementada con estaciones GPS permanentes localizadas a lo largo de la costa del Pacifico mexicano espaciadas, por lo menos, cada 65 km. Los datos de esta red GPS requieren ser transmitidos a una estacion central y procesados en tiempo real para monitorear la posicion de las estaciones. Asumiendo que lo anterior estuviera implementado, en este trabajo desarrollamos una metodologia para la estimacion rapida de parametros criticos de la fuente sismica y demostramos su viabilidad aplicandola al analisis del sismo de Colima-Jalisco de 1995 (M{sub w}=8.0) y de Sumatra-Andaman de 2004 (M{sub w}=9.0-9.3). Para este par de temblores, la metodologia propuesta arrojo valores de M{sub w} y E{sub s} que son muy similares a los reportados anteriormente, obtenidos a partir de analisis detallados. En el caso del sismo de Colima-Jalisco la localizacion estimada y el tamano del area de ruptura estan de acuerdo con los valores que se pueden inferir a partir de las localizaciones de las replicas. Actualmente existen 13 estaciones GPS permanentes a lo largo de la costa del Pacifico mexicano con un espaciamiento promedio de {approx}200 km, las cuales operan de manera autonoma. Es urgente incrementar este numero de estaciones a {>=}28 (logrando un espaciamiento de {<=}65 km), asi como transmitir los datos y determinar la posicion de las estaciones en tiempo real (preferiblemente cada segundo).

Pertussis toxin treatment differentially affects cholinergic and dopaminergic receptor stimulation of midbrain dopaminergic neurons.

Extracellular single-unit recordings and iontophoresis were used to compare the effect of a single administration of pertussis toxin (PTX, 1 microg), into midbrain dopamine (DA) nuclei (A9 and A10 regions), on the muscarinic, NMDA and DA receptor responses of midbrain DA cells in the anesthetized rat. Iontophoretic applications of DA, or apomorphine (50 microg/kg, i.v.), markedly reduced the firing of DA cells in control rats. In PTX-treated animals, these inhibitory responses were totally abolished, indicating that, in both DA nuclei, the inhibitory DA receptors are coupled to Gi/o proteins. In parallel, there was a significant decrease in the number of active DA cells per track which returned to baseline 5 weeks after the treatment. Applications of the muscarinic agonist oxotremorine M (OXO M) or of NMDA produced a potent increase in the firing of DA cells in control rats. DA neurons treated with PTX were still responsive to OXO M, although their sensitivity to the agonist was significantly reduced by 40%. In contrast, NMDA-induced activation remained unchanged, indicating that PTX did not non-selectively dampen all excitatory responses. Applications of cell-permeable cAMP derivatives did not change the basal firing of DA neurons. On the other hand, the phospholipase C inhibitors neomycin and ET-18-OCH3 (200 microg, i.c.v.), reduced significantly the activation of DA cells induced by OXO M. These data suggest that muscarinic activation of DA cells involves an M1-like receptor, possibly coupled to Gq/11 proteins, but also the participation of a PTX substrate.

Extracellular truncated influenza virus nucleoprotein.

In the culture medium of MDCK cells infected with influenza A/Duck/Ukraine/1/63(H3N8) virus two kinds of virus nucleoprotein (NP) are detected: full-length 56 kDa NP and truncated 53 kDa NP. However, in infected cells 53 kDa NP may be detected only at short pulse and after 10 min chase it becomes nondetectable. The extracellular truncated 53 kDa NP is detected in free RNP, and not in the virions. Both extracellular free 53 and 56 kDa NP in the virions are completely oligomerized. Several data argue against the possibility of extracellular 53 kDa NP formation being a result of extracellular 56 kDa NP proteolytic degradation. Thus, the accumulation of extracellular 53 kDa NP takes place only in the course of infection, and the amount of 53 kDa NP is not increased during prolonged storage of cell-free culture medium at +37 degrees C. Moreover, all extracellular 56 kDa NP of A/Duck/Ukraine/1/63 influenza virus is present in the oligomeric form, and the latter, in contrast to the mononeric form, is highly resistant to proteases. The possibility is discussed that in the course of A/Duck/Ukraine/1/63 (H3N8) influenza virus infection a fraction of the synthesized 56 kDa monomeric NP undergoes the proteolytic cleavage in the infected cells before oligomerization and forms the 53 kDa NP. This 53 kDa NP is then oligomerized, enters the RNP and is quickly secreted from the cells.

Effects of acute and repeated administration of salvinorin A on dopamine function in the rat dorsal striatum.

RATIONALE: Acute systemic administration of salvinorin A, a naturally occurring kappa-opioid receptor (KOPr) agonist, decreases locomotion and striatal dopamine (DA) overflow. OBJECTIVES: Conventional and quantitative microdialysis techniques were used to determine whether salvinorin A infusion into the dorsal striatum (DSTR) decreases DA overflow by altering DA uptake or release. The influence of repeated salvinorin A administration on basal DA dynamics and cocaine-evoked alterations in DA overflow and locomotion was also assessed. MATERIALS AND METHODS: Salvinorin A was administered via the dialysis probe (0; 20-200 nM) or via intraperitoneal (i.p.) injection (1.0 or 3.2 mg/kg per day x 5 days). The effects of a challenge dose of cocaine were examined 48 h after repeated salvinorin treatment. RESULTS: Retrodialysis of salvinorin A produced a dose-related, KOPr antagonist reversible, decrease in DA levels. Extracellular DA levels were decreased whereas DA extraction fraction, which provides an estimate of DA uptake, was unaltered. In contrast to its acute administration, repeated salvinorin A administration did not modify dialysate DA levels. Similarly, neither basal extracellular DA levels nor DA uptake was altered. Unlike synthetic KOPr agonists, prior repeated administration of salvinorin A did not attenuate the locomotor activating effects of an acute cocaine (20 mg/kg, i.p.) challenge. However, cocaine-evoked DA overflow was enhanced. CONCLUSIONS: These data demonstrate that acute, but not repeated, salvinorin A administration decreases mesostriatal neurotransmission and that activation of DSTR KOPr is sufficient for this effect. Differences in the interaction of salvinorin and synthetic KOPr agonists with cocaine suggest that the pharmacology of these agents may differ.

Darbepoietin alfa potentiates the efficacy of radiation therapy in mice with corrected or uncorrected anemia.

Darbepoietin alfa (DA) is a long-acting analogue of erythropoietin that has reduced receptor affinity and enhanced biological activity. Experiments were done to test the hypothesis that correction of anemia in tumor-bearing mice by DA would increase tumor oxygenation and potentiate radiation-induced tumor cell killing. A SCC VII tumor model was used to study tumor responses to fractionated radiation therapy in mice with anemia induced by total body irradiation. Administration of DA reduced the extent and duration of anemia and associated tumor hypoxia, protected the bone marrow cells and prevented the body weight loss from the effect of irradiation, and facilitated the recovery in a time-dependent manner, with the administration of DA prior to total body irradiation having the greatest protective effect. When combined with fractionated radiation therapy, DA increased the tumor growth delay time from 2.7 days for irradiation alone to 7.3 to 10.6 days for combination of DA and irradiation. The effect of DA on tumor responses to fractionated radiation therapy was observed when DA was given 18 to 4 days before starting radiation therapy, but DA was also equally effective as a radiosensitizer when given only 2 hours before fractionated irradiation therapy. Weekly dosing of DA was as efficacious for the enhancement of radiation responses of tumors as biweekly dosing. Similar results were obtained in the RIF-1 fibrosarcoma tumor model. These studies show that DA can effectively correct anemia in tumor-bearing mice and sensitize tumor cells to fractionated radiation therapy. Importantly, DA was also able to sensitize tumors to radiation in mice with uncorrected anemia and hypoxia, suggesting that the effect of DA on radiosensitivity was independent of these factors and a different mechanism of action may be responsible for this effect.

In this article, the characterization and evolution of pseudo-endoglucanases and a putative expansin-like gene in the migratory nematode Ditylenchus africanus are described. Four genes were cloned with a very high similarity to the endoglucanase Da-eng1, which, however, lack a part of the catalytic domain most probably due to homologous recombination. Owing to this deletion, at least one of the catalytic residues of the corresponding protein is missing, and hence these genes are possibly pseudogenes. Route of Nicotine Administration Influences In Vivo Dopamine Neuron Activity: Habituation, Needle Injection, and Cannula Infusion

Full Text Available.Mesolimbic dopamine (DA) systems play a critical role in tobacco addiction driven by nicotine. Nicotine activates midbrain DA neurons and, consequently, elevates DA concentrations in targets, especially in the nucleus accumbens (NAc) of the ventral striatum. The route of drug administration influences the impact of addictive drugs. Here, we examine whether the nature of the administration alters DA neuron activity and DA concentrations in the NAc. Using unhabituated naïve freely moving rats, microdialysis measurements showed that nicotine administered via needle injection caused greater DA release in the NAc than the same dose administered via an implanted chronic cannula. After habituation to the needle injections, however, there was no significant difference in DA signaling between the needle and cannula routes of administration. Consistent with these microdialysis results after habituation, our in vivo tetrode unit recordings showed no significant difference in midbrain DA neuron activity in response to nicotine delivered by needle or cannula as long as predictive cues were avoided.

USF is a cellular factor involved in the transcriptional regulation of several cellular and viral promoters. Purified USF HeLa cells (HeLA USF) consists of 43- and 44-kDa polypeptides which show independent binding to a specific DNA element. A cDNA encoding the 43-kDa species has been previously cloned. The authors here that the purified form of bacterially expressed 43-kDa USF (1) exists in solution as a dimer whose formation is greatly favored under reducing conditions, (2) binds to its cognate DNA sequence in a manner indistinguishable from that of HeLa USF, and (3) is as efficient as HeLa USF in stimulating transcription from target is immunologically unrelated to the 43-kDa polypeptide but is associated with it in HeLa cell extracts. These results suggests that the 43-kDa component possesses an intrinsic DNA binding the transcriptional activation potential and that the 44-kDa USF component of the natural USF complex may have some regulatory role.

(3H)spiperone (SPI) binding in vivo, biochemical parameters and behavior were measured after modulating DA levels by various drug treatments. DA releasers and uptake inhibitors increased SPI binding in rat striatum. In other brain areas, the effects were variable, but only the pituitary remained unaffected. Surprisingly, nomifensine decreased SPI binding in frontal cortex. The effects of these drugs were monitored by measuring DA, serotonin (5-HT) and their metabolites in the same rats. The increased SPI binding in striatum was parallel to the locomotor stimulation with the following rank order: amfonelic acid greater than nomifensine greater than D-amphetamine greater than or equal to methylphenidate greater than amineptine greater than bupropion. Decreasing DA levels with reserpine or alpha-methyl-para-tyrosine reduced SPI binding by 45% in striatum only when both drugs were combined. In contrast, reserpine enhanced SPI binding in pituitary. Thus, the amount of releasable DA seems to modulate SPI binding characteristics. It is suggested that in vivo, DA receptors are submitted to dynamic regulation in response to changes in intrasynaptic concentrations of DA.

This work presents the authors experience on the viability of FURNAS partnership with the Nacional Energetica for completion of the Serra da Mesa power plant. The partnership refers to the transmission system use for distribution of 51.54 % of the partner power to various consumers in different points of the system. Special emphasis is placed on the evaluation of the transactions with LIGHT-R J and the Brasilia electric company.

In intestinal microvilli, the 110K-calmodulin complex is the major component of the cross-bridges which connect the core bundle of actin filaments to the membrane. The authors previous work showed that the 110-kDa polypeptide can be divided into three functional domains: a 78-dDa fragment that contains the ATPase activity and the ATP-reversible F-actin-binding site, a 12-kDa fragment required for binding calmodulin molecules, and a terminal 20-kDa domain of unkown function. By analysis of limited {alpha}-chymotryptic cleavage products, they now show that the molecular organization is very similar to that described for the S{sub 1} fragment of myosin. The catalytic site was identified by photoaffinity labeling with (5,6-{sup 3}H)UTP, and fragments binding F-actin were identified by cosedimentation assays. Cleavage of the 78-kDa fragment yielded major fragments of 32 and 45 kDa, followed by cleavage of the 45-kDa fragment to a 40-kDa fragment. Of these, only the 32-kDa fragment was labeled by (5,6-{sup 3}H)UTP. They conclude that the catalytic site is located in the 32-kDa fragment and the F-actin-binding site is present in the 45-kDa fragment; the ability to bind actin is lost upon further cleavage of the 45-kDa fragment to 40 kDa. Peptide sequence analysis revealed that the 45-kDa fragment lies within the molecule and suggests that the 32-kDa fragment is the amino terminus. These results suggest that the 12-kDa calmodulin-binding region confers a Ca{sup 2{plus}}-dependent accessibility to the F-actin-protectable cleavage site in the S{sub 1}-like domain. This is the first evidence for a Ca{sup 2{plus}}-regulated structural change in the S{sub 1}-like domain of the 110K-calmodulin complex.
