The necrogenic bacterium Erwinia amylovora responsible for the fire blight disease causes cell death in apple tissues to enrich intercellular spaces with nutrients. Apple leaves contain large amounts of dihydrochalcones (DHCs), including phloridzin and its aglycone phloretin. Previous work showed an important decrease in the constitutive DHCs stock in infected leaves, probably caused by transformation reactions during the infection process. At least two flavonoid transformation pathways have been described so far: deglucosylation and oxidation. The aim of the present study was to determine whether DHCs are differentially converted in two apple genotypes displaying contrasted susceptibilities to the disease. Different analyses were performed: i) enzymatic activity assays in infected leaves, ii) identification/quantification of end-products obtained after in vitro enzymatic reactions with DHCs, iii) evaluation of the bactericidal activity of end-products. The results of the enzymatic assays showed that deglucosylation was dominant over oxidation in the susceptible genotype MM106 while the opposite was observed in the resistant genotype Evereste. These data were confirmed by LC-UV/Vis-MS analysis of in vitro reaction mixtures, especially because higher levels of o-quinoid oxidation products of phloretin were measured by using the enzymatic extracts of Evereste infected leaves. Their presence correlated well with a strong bactericidal activity of the reaction mixtures. Thus, our results suggest that a differential transformation of DHCs occur in apple genotypes with a potential involvement in the establishment of the susceptibility or the resistance to fire blight, through the release of glucose or of highly bactericidal compounds respectively. The involvement of choline and its metabolite trimethylamine-N-oxide (TMAO) in endothelial dysfunction and atherosclerosis has been repeatedly confirmed. Phloretin, a dihydrochalcone flavonoid usually present in apples, possesses a variety of biological activities including vascular nutrition. This study was designed to investigate whether phloretin could alleviate or prevent high choline-induced vascular endothelial dysfunction and liver injury in mice. Mice were provided with 3% high choline water and given phloretin orally daily for 10 weeks. The high choline-treated mice showed the significant dyslipidemia and hyperglycemia with the impaired liver and vascular endothelium (p < 0.01). Administration of phloretin at 200 and 400 mg/kg bw significantly reduced the choline-induced elevation of serum TC, TG, LDL-C, AST, ALT, ET-1 and TXA2 (p < 0.01), and markedly antagonized the choline-induced decrease of serum PGI2, HDL-C and NO levels. Furthermore, phloretin elevated hepatic SOD and GSH-Px activities and decreased hepatic MDA levels of the mice exposed to high choline water. Moreover, histopathological test with the H&E and Oil Red O staining of liver sections confirmed the high choline diet-caused liver steatosis and the hepatoprotective effect of phloretin. These findings suggest that high choline causes oxidative damage, and phloretin alleviate vascular endothelial dysfunction and liver injury. PMID:27316781

Full Text Available The antioxidant properties of two apple dihydrochalcones, namely phloretin and phloridzin, were evaluated and compared with those of α-tocopherol and butylated hydroxytoluene (BHT. The effects were studied in an oil-in-water emulsion system containing methyl linolenate (ML, methyl eicosapentaenoate (MEPA, and methyl docosahexaenoate (MDHA in which oxidation was initiated by the peroxyl radical generator 2,2-azobis(2-amidinopropane dihydrochloride (AAPH and in fish oil where oxidation was initiated thermally. In the emulsion system, phloretin (1 and 5 mM completely inhibited the oxidation of ML tested as evidenced by the thiobarbituric acid reactive substances (TBARS assay. Under the same conditions, phloridzin was less effective than phloretin, but still more effective than α-tocopherol. Both phloretin and phloridzin molecules had a marginal inhibitory effect against oxidation of fish oil induced by heating at 70 °C for 3 hours, when compared to BHT. These results indicate that phloretin and phloridzin have the potential to suppress lipid oxidation in polyunsaturated fatty acid (PUFA containing foods.

We hypothesised that consumption of flavanol-containing apple puree would modulate platelet activity and increase nitric oxide metabolite status, and that high flavanol apple puree would exert a greater effect than low flavanol apple puree. 25 subjects consumed 230 g of apple puree containing 25 and 100mg epicatechin (low and high flavanol apple puree, respectively) and aspirin (75 mg) in random order. Measurements were made at baseline, acutely after treatment (2, 6 and 24 h), and after 14 d of treatment. Low flavanol apple puree significantly attenuated ADP and epinephrine-induced integrin-β3 expression 2 h and 6 h after consumption and ADP and epinephrine-induced P-selectin expression within 2h of consumption. High flavanol apple puree attenuated epinephrine and ADP-induced integrin-β3 expression after 2 and 6h. ADP and epinephrine-induced integrin-β3 expression was significantly attenuated 2, 6 and 24 h after consumption of aspirin, whilst 14 d aspirin consumption attenuated collagen-induced P-selectin expression only. The plasma total nitric oxide metabolite conc. was significantly increased 6h after consumption of both low and high flavanol apple purees. In conclusion, consumption of apple purees containing ⩾25 or 100 mg flavanols transiently attenuated ex vivo integrin-β3 and P-selectin expression and increased plasma nitric oxide metabolite conc. in healthy subjects, but the effect was not enhanced for the high flavanol apple puree. Some Alternaria species are able to produce plant pathogenic as well as toxic metabolites. In both agriculture and the food industry it is important know if toxigenic Alternaria are present to rapidly employ the correct corrective actions. The purpose of this work was to establish a real-time PCR method, which can detect and quantify apple pathogenic and toxigenic Alternaria. An AM-toxin I primer set, which could recognize Alternaria DNA only, was designed by using primers complementary to the AM-toxin I gene. The method could detect small amounts of DNA (4 pg) and still obtain a large dynamic range (4 decades) without interference from apple material. Eight Alternaria isolates were analyzed for the presence of AM-toxin I gene and their production of secondary metabolites. Then analyses showed that all eight isolates contained the AM toxin gene and were able to produce the plant pathogenic tentoxin in addition to AM toxin I. The analyses also showed the production of tenuazonic acid, alternariols, Altenuene, altenusin and/or altertoxin I in pure culture. Analyses of inoculated apples showed that both the AM-toxin gene and alternariol monomethyl ether could be detected. Morphological analyses suggested that the eight Alternaria strains, though they all carried the AM toxin genes, probably belong to different but closely related un-described Alternaria taxa in the A. tenuissima species-group based on morphological and chemical differences. Urea is actively transported inwardly (Ji) across the skin of the green toad Bufo viridis. Ji is markedly enhanced in toads adapted to hypertonic saline. The authors studied urea transport across the skin of Bufo viridis under a variety of experimental conditions, including treatment with amiloride and phloretin, agents that inhibit urea permeability in the bladder of Bufo marinus. Amiloride (10-4 M) significantly inhibited Ji in both adapted and unadapted animals and was unaffected by removal of sodium from the external medium. Phloretin (10-4 M) significantly inhibited Ji in adapted animals by 23-46%; there was also a reduction in Ji in unadapted toads at 10-4 and 5 x 10-4 M phloretin. A dose-response study revealed that the concentration of phloretin causing half-maximal inhibition (K1/2) was 5 x 10-4 M for adapted animals. Ji was unaffected by the substitution of sucrose for Ringer solution or by ouabain. They conclude (1) the process of adaptation appears to involve an increase in the number of amiloride- and phloretin-inhibitable urea transport sites in the skin, with a possible increase in the affinity of the sites for phloretin; (2) the adapted skin resembles the Bufo marinus urinary bladder with respect to amiloride and phloretin-inhibitable sites; (3) they confirm earlier observations that Ji is independent of sodium transport

The shaded peel of 'Fortune' (a red cultivar) and 'Mutsu' (a yellow/green cultivar) apple (Malus domestica Borkh.) was exposed to full sun by turning fruit 180° at about one week before harvest to determine the expression of key genes involved in anthocyanin synthesis in response to sunlight exposure and their relationships with the levels of anthocyanins and other phenolics. For the unturned (control) fruit, the shaded peel had lower expression levels of MdMYB10 (a transcriptional factor in the regulation of anthocyanin biosynthesis) and seven structural genes in anthocyanin synthesis (MdPAL, MdCHS, MdCHI, MdF3H, MdDFR1, MdLDOX, and MdUFGT), and lower levels of anthocyanins and flavonols than the sun-exposed peel in both cultivars. Exposure of the shaded peel to full sun caused marked up-regulation of the expression of MdMYB10 and all seven structural genes, which peaked between 6 h and 30 h after fruit turning, consequently leading to higher levels of anthocyanins, flavonols, and total phenolics than in the shaded peel and even in the sun-exposed peel of control fruit. Interestingly, the levels of flavonols were higher in the shaded peel of turned fruit (the original sun-exposed peel) than in the sun-exposed peel of both control and turned fruit in both cultivars, suggesting that competition for substrates exists in different branches of the phenylpropanoid pathway. These results indicate that sunlight exposure stimulates the expression of MdMYB10 and structural genes in anthocyanin synthesis, thereby elevating the levels of anthocyanins and other phenolic compounds in both red and yellow/green cultivars. 目的：建立基于 QuEChERS-高效液相色谱-串联质谱测定苹果中的丁醚脲及其代谢物丁醚脲-脲和丁醚脲-甲酰胺残留量的方法。方法样品经乙腈提取后用PSA作为固相分散萃取剂净化除杂，以0.1%甲酸乙腈溶液和0.1%甲酸水溶液(含10 mmol/L甲酸铵)为流动相进行梯度洗脱，高效液相色谱-串联质谱法正离子多反应监测模式(MRM)测定。结果丁醚脲及其代谢物在5~100μg/L的范围内有良好的线性，相关系数(R2)>0.999。定量限5μg/kg为以空白苹果为基质，在5、10、50μg/kg三个水平的加标回收率范围在78.3%~103.1%之间，精密度在2.1%~8.2%之间。结论该方法前处理快速简单，可用于对进出口的苹果中丁醚脲及其代谢物的检测。%ABSTRACT:ObjectiveTo establish a method to detect the diafenthiuron and its metabolites residue in apple by QuEChERS-high performance liquid chromatography-tandem mass spectrometry.Methods The samples were extracted with acetonitrile and cleaned up by PSA. The separation was carried out with a gradient elution using 0.1% formic acid acetonitrile solution and 0.1% formic acid solution (10 mmol/L ammonium formate) as mobile phases. The analysis of diafenthiuron and its metabolites was performed under electrospray positive ionization MRM mode.Results A good linearity (R2>0.999) was achieved for the target compounds in the range of 5~100μg/L. The limit of quantification was 5μg/kg. The recoveries at the three spiked levels (5, 10, and 20μg/kg) in the blank matrix were ranged from 78.3% to103.1%, with the relative standard deviations varied from 2.1% to 8.2%. ConclusionThe method is accurate and efficient, and can be applied to the determination of diafenthiuron and its metabolites residue in apple for import and export.

Flavors of "Fuji" apple cultivated with or without synthetic agrochemicals were compared using quantitative descriptive analyses (QDA) and metabolite profiling for 3 seasons. Experimental plots included conventional crops (with agrochemicals) and organic crops (without agrochemicals) at our institute and organic and conventional farms. Additionally, mass market samples were analyzed. Organic apples were weak in sweetness and floral characteristics and had enhanced green and sour flavors. Most esters and sugars were present in lower concentrations in organic than in conventional apples. Close relation of principal component 1 of QDA and metabolite profiles, to ethylene production suggested that ethylene is considerably involved in flavor synthesis. Reduced ethylene associated with immaturity accounted for insufficient flavor synthesis and weak aroma and flavor attributes of organic apples. Furthermore, organic apples from the farm were more flavorsome than those from the institute in 2012, suggesting possible recovery of ethylene production after a long organic cultivation period. Full Text Available Evidence suggests that a diet high in fruits and vegetables may decrease the risk of chronic diseases, such as cardiovascular disease and cancer, and phytochemicals including phenolics, flavonoids and carotenoids from fruits and vegetables may play a key role in reducing chronic disease risk. Storage has little to no effect on apple phytochemicals, but processing can greatly affect apple phytochemicals. Apple Valsa canker, caused by the fungus Valsa mali (Vm), is one of the most destructive diseases of apple in China. A better understanding of this host-pathogen interaction is urgently needed to improve management strategies. In the current study we sequenced the transcriptomes of Vm during infection of apple bark and mycelium grown in axenic culture using Illumina RNA-Seq technology. We identified 437 genes that were differentially expressed during fungal infection compared to fungal mycelium grown in axenic culture. One hundred and thirty nine of these 437 genes showed more than two fold higher transcript abundance during infection. GO and KEGG enrichment analyses of the up-regulated genes suggest prevalence of genes associated with pectin catabolic, hydrolase activity and secondary metabolite biosynthesis during fungal infection. Some of the up-regulated genes associated with loss of pathogenicity and reduced virulence annotated by host-pathogen interaction databases may also be involved in cell wall hydrolysis and secondary metabolite transport, including a glycoside hydrolase family 28 protein, a peptidase and two major facilitator superfamily proteins. This highlights the importance of secondary metabolites and cell wall hydrolases during establishment of apple Valsa canker. Functional verification of the genes involved in pathogenicity of Vm will allow us to better understand how the fungus interferes with the host machinery and assists in apple canker establishment. Of two stereoisomers of glucose, only D- and not L-glucose is abundantly found in nature, being utilized as an essential fuel by most organisms. The uptake of D-glucose into mammalian cells occurs through glucose transporters such as GLUTs, and this process has been effectively monitored by a fluorescent D-glucose derivative 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-D-glucose (2-NBDG) at the single cell level. However, since fluorescence is an arbitrary measure, we have developed a fluorescent analog of L-glucose 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-L-glucose (2-NBDLG), as a negative control substrate for more accurately identifying the stereoselectivity of the uptake. Interestingly, a small portion of mouse insulinoma cells MIN6 abundantly took up 2-NBDLG at a late culture stage (≳ 10 days in vitro, DIV) when multi-cellular spheroids exhibiting heterogeneous nuclei were formed, whereas no such uptake was detected at an early culture stage (≲ 6 DIV). The 2-NBDLG uptake was persistently observed in the presence of a GLUT inhibitor cytochalasin B. Neither D- nor L-glucose in 50 mM abolished the uptake. No significant inhibition was detected by inactivating sodium/glucose cotransporters (SGLTs) with Na(+)-free condition. To our surprise, the 2-NBDLG uptake was totally inhibited by phloretin, a broad spectrum inhibitor against transporters/channels including GLUTs and aquaporins. From these, a question might be raised if non-GLUT/non-SGLT pathways participate in the 2-NBDLG uptake into spheroid-forming MIN6 insulinoma. It might also be worthwhile investigating whether 2-NBDLG can be used as a functional probe for detecting cancer, since the nuclear heterogeneity is among critical features of malignancy. Full Text Available Volatile organic compounds (volatiles comprise a chemically diverse class of low molecular weight organic compounds having an appreciable vapor pressure under ambient conditions. Volatiles produced by plants attract pollinators and seed dispersers, and provide defense against pests and pathogens. For insects, volatiles may act as pheromones directing social behavior or as cues for finding hosts or prey. For humans, volatiles are important as flavorants and as possible disease biomarkers. The marine environment is also a major source of halogenated and sulfur-containing volatiles which participate in the global cycling of these elements. While volatile analysis commonly measures a rather restricted set of analytes, the diverse and extreme physical properties of volatiles provide unique analytical challenges. Volatiles constitute only a small proportion of the total number of metabolites produced by living organisms, however, because of their roles as signaling molecules (semiochemicals both within and between organisms, accurately measuring and determining the roles of these compounds is crucial to an integrated understanding of living systems. This review summarizes recent developments in volatile research from a metabolomics perspective with a focus on the role of recent technical innovation in developing new areas of volatile research and expanding the range of ecological interactions which may be mediated by volatile organic metabolites.

Root Learning, a learning consulting organization with a background in strategic planning, recognizes the knowledge gap that frequently exists between a leadership team and the rest of an organization. Team members supposedly working toward the same goal don't always have the same vision as to where the organization is headed, and they may not understand how the piece they are accountable for fits into the big picture. To address these complex problems, Root Learning utilizes the age-old tools of sarcasm, metaphor and graphics (much in the same way that ASK uses a traditional storytelling format.) Full Text Available Aqrobacterium tumefaciens and cucumber, mustard and linseeds were compared as test organisms for evaluation of the biological activity of patulin. It was found that the reaction of cucumber seeds and linseed to the patulin concentrations was more pronounced than that of mustard and Aqrobacterium tumefaciens. The activity of metabolites produced by Penicillium expansum was investigated with the use of cucumber seeds. As measure of activity served the percentage of radicule growth inhibition was compared with the growth in control seeds. The biological activity of the metabolites was specific for the isolates, those from apples being more active. Thirty two isolates from pears and 34 from apples were examined.

Alternaria core rot is a major postharvest disease of apple fruit in several countries of the world, including Greece. The study was conducted aiming to identify the disease causal agents at species level, investigate the aggressiveness of Alternaria spp. isolates and the susceptibility of different apple varieties and determine the mycotoxigenic potential of Alternaria spp. isolates from apple fruit. Seventy-five Alternaria spp. isolates obtained from apple fruit showing core rot symptoms were identified as either Alternaria tenuissima or Alternaria arborescens at frequencies of 89.3 and 11.7%, respectively, based on the sequence of endopolygalacturonase (EndoPG) gene. Artificial inoculations of fruit of 4 different varieties (Fuji, Golden Delicious, Granny Smith and Red Delicious) and incubation at two different temperatures (2 and 25°C) showed that fruit of Fuji variety were the most susceptible and fruit of Golden Delicious the most resistant to both pathogens. In addition, the production of 3 mycotoxins, alternariol (AOH), alternariol monomethyl ether (AME) and tentoxin (TEN) was investigated in 30 isolates of both species. Mycotoxin determination was conducted both in vitro, on artificial nutrient medium and in vivo on artificially inoculated apple fruit, using a high performance liquid chromatography with diode array detector (HPLC-DAD). The results showed that most of the isolates of both species were able to produce all the 3 metabolites both in vivo and in vitro. On apple fruit A. tenuissima isolates produced more AOH than A. arborescens isolates, whereas the latter produced more TEN than the former. Such results indicate that Alternaria core rot represents a major threat of apple fruit production not only due to quantitative yield losses but also for qualitative deterioration of apple by-products. Full Text Available Proanthocyanidins (PAs are the major component of phenolics in apple, but mechanisms involved in PA biosynthesis remain unclear. Here, the relationship between the PA biosynthesis and the expression of genes encoding leucoanthocyanidin reductase (LAR and anthocyanidin reductase (ANR was investigated in fruit skin of one apple cultivar and three crabapples. Transcript levels of LAR1 and ANR2 genes were significantly correlated with the contents of catechin and epicatechin, respectively, which suggests their active roles in PA synthesis. Surprisingly, transcript levels for both LAR1 and LAR2 genes were almost undetectable in two crabapples that accumulated both flavan-3-ols and PAs. This contradicts the previous finding that LAR1 gene is a strong candidate regulating the accumulation of metabolites such as epicatechin and PAs in apple. Ectopic expression of apple MdLAR1 gene in tobacco suppresses expression of the late genes in anthocyanin biosynthetic pathway, resulting in loss of anthocyanin in flowers. Interestingly, a decrease in PA biosynthesis was also observed in flowers of transgenic tobacco plants overexpressing the MdLAR1 gene, which could be attributed to decreased expression of both the NtANR1 and NtANR2 genes. Our study not only confirms the in vivo function of apple LAR1 gene, but it is also helpful for understanding the mechanism of PA biosynthesis.

Full Text Available Patulin is a mycotoxin produced by several species of Penicillium, Aspergillus and Byssochlamys. In nature, it is found diffusely at various products and also in apple and apple products. Because of the negative effects of patulin on human health, ıt ıs started to be used as an important quality parameter especially in apple juice and various apple products. On the other hand, having water solubility and stability to the heat treatment properties acquired another importance to the patulin in apple juice and apple juice concentrate. Removal of rotten parts of apples that will be processed to the apple juice and apple juice concentrate made the end product reliable in view of patulin. At apple juice and apple juice concentrate factories, activated charcoal is used diffusely for the removal of patulin passed to the apple juice. Exterior of activated charcoal, with the addition of ascorbic acid and/or sorbates, sulfhydryl (SH components or different food ingredients (cinnamon oil, potassium sorbate e.t.c and treatment of low dose radiation and modified atmosphere to the apple juice were being effective at the reduction of patulin. There are contradictory results about the inactivation of patulin with heat treatment and storage.

Full Text Available The new apple cultivar �Sauron� was obtained at Fruit Research Station Cluj, Romania (FRS Cluj, and homologated in 2007. �Sauron� was identified in open pollinated population of �Cluj 3/83� selection [�Cluj III-VI-5-26� selection (�Parmain d�Or�, open pollinated x �NJ 46�]. The trees have moderate vigour, upright then spread, with moderate productivity. Fruits are medium to large, usually red, with a portion being greenish or yellow-green and purple red vertically striped. The fruit has good quality, being soft eating apple due to their lack of crispness. Quality indices include firmness, crispness and excellent flavour. Fruits become ripe between 15 August and 5 September and the fruits are good quality, being proper for dessert and industrialization (juice, applesauce, pies, and cider.

In the focus of the first part of this work was the modulation of the promoter methylation of the tumor suppressor gene p16INK4a by an extract of Bohnapfel (BAE) as well as its main constituent n-chlorogenic acid (CGA) in both, human colon carcinoma cells (HT29) and during a pilot intervention trial with type-II diabetic patients. BAE as well as CGA partially displayed a decrease in DNA methylation of the p16INK4a promoter after 48 h and 96 h incubation, however not in a time- and concentration-dependent manner. In addition, none of the two test compounds showed a re-activation of the tumor suppressor on both, transcription and protein levels. In a pilot trial with type-II diabetics 2 of 15 subjects reflected a methylation of the p16INK4a promoter in exfoliated colonocytes of human feaces. After four weeks of intervention with cloudy apple juice a reduction of the p16INK4a promoter methylation was observed in these two subjects. The focus of the second section of this thesis was the question whether polyphenol-rich apple pomace extracts (AT), obtained from depectinized pomace, are able to affect the intracellular redox state of HT29 cells allowing the classification of their potency compared to similarly prepared extracts of apple juice (AS). The studies showed that AT possesses a significantly higher bioactivity than AS regarding the reduction of intracellular ROS as well as the increase of the transcription of the enzyme glutamylcysteinligase (GCL). Further investigations indicated an activation of the Nrf2-ARE pathway and a downstream induction of selected enzymes of the phase-II metabolism. Apple varieties of Guoguang, Fuji, Hongxiangjiao and Huangxiangjiao were used as materal, the quality of fresh apple and explosion puffed products were analyzed based on the data of their physical and chemical characteristics and flavor. The results showed that Guoguang apple had the optimal flavor and Huangxiangjiao apple had the optimal product quality. Full Text Available Four apples varieties (Braebum, Elstar, Jonagold and Teser T219 were tested to study techniques for instrumental measurement of the mealiness of apple fruit. The measurements were done just after cold storage (2Ã‚Â°C, 95% relative humidity and repeated one week of storage at 20 Ã‚Â°C, 65% relative humidity later. The non-destructive acoustic response technique was used as a measure of fruit firmness. Texture profile analysis (TPA and tensile tests were applied to measure the hardness, cohe-siveness and adhesion of the apple tissue. The juiciness, soluble solids content (SSC, pH value and internal air space (IAS of the apples were determined. The results show that the TPA technique and tensile test may be derived as mechanical ways to measure apple mealiness. Apple fruit internal air space increases and juiciness decreases as the apple becomes mealy, and they may be good indications of mealiness. The mealiness criteria may be different for different varieties of apples.

Full Text Available The aim of this work was to evaluate the nitrogen content in different varieties of apple musts and to study the effect of different nitrogen concentrations in apple wine fermentation. The average total nitrogen content in 51 different apples juices was 155.81 mg/L, with 86.28 % of the values above 100 mg/L. The apple must with 59.0, 122.0 and 163.0 mg/L of total nitrogen content showed the maximum population of 2.05x 10(7; 4.42 x 10(7 and 8.66 x 10(7 cell/mL, respectively. Therefore, the maximum fermentation rates were dependent on the initial nitrogen level, corresponding to 1.4, 5.1 and 9.2 g/L.day, respectively. The nitrogen content in the apple musts was an important factor of growth and fermentation velocity.

Carotenoid accumulation confers distinct colouration to plant tissues, with effects on plant response to light and as well as health benefits for consumers of plant products. The carotenoid pathway is controlled by flux of metabolites, rate-limiting enzyme steps, feed-back inhibition, and the strength of sink organelles, the plastids, in the cell. In apple (Malus × domestica Borkh), fruit carotenoid concentrations are low in comparison with those in other fruit species. The apple fruit flesh, in particular, begins development with high amounts of chlorophylls and carotenoids, but in all commercial cultivars a large proportion of this is lost by fruit maturity. To understand the control of carotenoid concentrations in apple fruit, metabolic and gene expression analysis of the carotenoid pathway were measured in genotypes with varying flesh and skin colour. Considerable variation in both carotenoid concentrations and compound profile was observed between tissues and genotypes, with carotenes and xanthophylls being found only in fruit accumulating high carotenoid concentrations. The study identified potential rate-limiting steps in carotenogenesis, which suggested that the expression of ZISO, CRTISO, and LCY-ε, in particular, were significant in predicting final carotenoid accumulation in mature apple fruit. Full Text Available Over the last 60 years, planting densities for apple have increased as improved management systems have been developed. Dwarfing rootstocks have been the key to the dramatic changes in tree size, spacing and early production. The Malling series of dwarfing rootstocks (M.9 and M.26 have been the most important dwarfing rootstocks in the world but are poorly adapted in some areas of the world and they are susceptible to the bacterial disease fire blight and the soil disease complex, apple replant disease which limits their uses in some areas. Rootstock breeding programs in several parts of the world are developing improved rootstocks with resistance to fire blight, and replant disease, and improved cold hardiness and yield efficiency. A second important trend has been the increasing importance of new cultivars. New cultivars have provided opportunities for higher prices until they are over-produced. A new trend is the "variety club" in which variety owners manage the production and marketing of a new unique cultivar to bring higher prices to the growers and variety owners. This has led to many fruit growers being unable to plant or grow some new cultivars. Important rootstock and cultivar genes have been mapped and can be used in marker assisted selection of future rootstock and cultivar selections. Other important improvements in apple culture include the development of pre-formed trees, the development of minimal pruning strategies and limb angle bending which have also contributed to the dramatic changes in early production in the 2nd-5th years after planting. Studies on light interception and distribution have led to improved tree forms with better fruit quality. Full Text Available The apple cultivar �Saruman� was obtained at Fruit Research Station Cluj, Romania (FRS Cluj, and homologated in 2007. �Saruman� was released by hybridization between �Cluj III-VI-5-26� selection (�Parmain d�Or�, open pollinated and �NJ 46�. The trees are vigorous, spreading shape, and with medium crop yield. The fruits have large size, conic shape and mostly red (purple coloration; they have white flesh with a sweet, crisp, aromatic flavour and low acidity. Fruits become ripe in the last decade of August, first decade of September and the fruits are proper for dessert and well suited for cooking, applesauce, cider, pies.

For food production in space at recycling bio-elements under closed environment, appropriate organisms should be chosen to drive the closed materials recycle loop. We propose a combination of green algae, photosynthetic protozoa, and aquatic plants such as Wolffia spp., for the primary producer fixing solar energy to chemical form in biomass, and apple snail, Pomacea bridgesii, which converts this biomass to animal meat. Because of high proliferation rate of green algae or protozoa compared to higher plants, and direct conversion of them to apple snail, the efficiency of food production in this combination is high, in terms of energy usage, space for rearing, and yield of edible biomass. Furthermore, green algae and apple snail can form a closed ecological system with exchanging bio-elements between two member, i.e. excreta of snail turn to fertilizer of algae, and grown algae become feed for snail. Since apple snail stays in water or on wet substrate, control of rearing is easy to make. Mass production technology of apple snail has been well established to utilize it as human food. Nutrients of apple snail are also listed in the standard tables of food composition in Japan. Nutrients for 100 g of apple snail canned in brine are energy 340 kJ, protein 16.5 g, lipid 1.0 g, cholesterol 240 mg, carbohydrate 0.8 g, Ca 400 mg, Fe 3.9 mg, Zn 1.5 mg. It is rich in minerals, especially Ca and Fe. Vitamin contents are quite low, but K 0.005 mg, B2 0.09 mg, B12 0.0006 mg, folate 0.001 mg, and E 0.6 mg. The amino acid score of apple snail could not be found in literature. Overall, apple snail provides rich protein and animal lipid such as cholesterol. It could be a good source of minerals. However, it does not give enough vitamin D and B12 , which are supposed to be supplemented by animal origin foods. Full Text Available Abstract Background Our study was part of the large European project ISAFRUIT aiming to reveal the biological explanations for the epidemiologically well-established health effects of fruits. The objective was to identify effects of apple and apple product consumption on the composition of the cecal microbial community in rats, as well as on a number of cecal parameters, which may be influenced by a changed microbiota. Results Principal Component Analysis (PCA of cecal microbiota profiles obtained by PCR-DGGE targeting bacterial 16S rRNA genes showed an effect of whole apples in a long-term feeding study (14 weeks, while no effects of apple juice, purée or pomace on microbial composition in cecum were observed. Administration of either 0.33 or 3.3% apple pectin in the diet resulted in considerable changes in the DGGE profiles. A 2-fold increase in the activity of beta-glucuronidase was observed in animals fed with pectin (7% in the diet for four weeks, as compared to control animals (P Bacteroidetes, whereas bands that became more prominent represented mainly Gram-positive anaerobic rods belonging to the phylum Firmicutes, and specific species belonging to the Clostridium Cluster XIVa. Quantitative real-time PCR confirmed a lower amount of given Bacteroidetes species in the pectin-fed rats as well as in the apple-fed rats in the four-week study (P Clostridium coccoides (belonging to Cluster XIVa, as well as of genes encoding butyryl-coenzyme A CoA transferase, which is involved in butyrate production, was detected by quantitative PCR in fecal samples from the pectin-fed animals. Conclusions Our findings show that consumption of apple pectin (7% in the diet increases the population of butyrate- and β-glucuronidase producing Clostridiales, and decreases the population of specific species within the Bacteroidetes group in the rat gut. Similar changes were not caused by consumption of whole apples, apple juice, purée or pomace.

Full Text Available Apple is economically the most important species of genus Malus Miller. In respect of production, trade and consumption, it ranks first among deciduous fruit and third on a global scale among all fruit species. Apple breeding is carried out on a large scale in several scientific institutes throughout the world. Due to this activity, apple is a fruit species with the highest number of described monogenic traits; 76 genes, encoding morphological traits, pest and disease resistance, as well as 69 genes encoding enzymes. The development of molecular markers (RFLPs, AFLPs, SCARs and SSRs has allowed the mapping of the apple genome and the development of several saturated genetic maps, to which genes controlling important traits are assigned. Markers flanking these genes not only play an important role in selecting parental combinations and seedlings with positive traits, but they are also particularly important in detecting recessive traits, such as seedless fruit. In addition they enable pre-selection for polygenic quantitative traits. In recent years, particular attention has been paid to biochemical and physiological processes involved in the pathway of important traits e.g., ripening and the storage capability of apple fruit.

Apple is a fruit crop of significant economic importance, and breeders world wide continue to develop novel cultivars with improved characteristics. The lengthy juvenile period and the large field space required to grow apple populations have imposed major limitations on breeding. Various molecular biological techniques have been employed to make apple breeding easier. Transgenic technology has facilitated the development of apples with resistance to fungal or bacterial diseases, improved fruit quality, or root stocks with better rooting or dwarfing ability. DNA markers for disease resistance (scab, powdery mildew, fire-blight, Alternaria blotch) and fruit skin color have also been developed, and marker-assisted selection (MAS) has been employed in breeding programs. In the last decade, genomic sequences and chromosome maps of various cultivars have become available, allowing the development of large SNP arrays, enabling efficient QTL mapping and genomic selection (GS). In recent years, new technologies for genetic improvement, such as trans-grafting, virus vectors, and genome-editing, have emerged. Full Text Available The aims of this study were to use the isotope analysis method to quantify the carbon of C3 photosynthetic cycle in commercial apple nectars and to determine the legal limit to identify the beverages that do not conform to the safety standards established by the Brazilian Ministry of Agriculture, Livestock and Food Supply. These beverages (apple nectars were produced in the laboratory according to the Brazilian legislation. Adulterated nectars were also produced with an amount of pulp juice below the permitted threshold limit value. The δ13C values of the apple nectars and their fractions (pulp and purified sugar were measured to quantify the C3 source percentage. In order to demonstrate the existence of adulteration, the values found were compared to the limit values established by the Brazilian Law. All commercial apple nectars analyzed were within the legal limits, which enabled to identify the nectars that were in conformity with the Brazilian Law. The isotopic methodology developed proved efficient to quantify the carbon of C3 origin in commercial apple nectars.

This study was exploratory in nature, with emphasis on initial purchases and not repeat purchases or long-term loyalties to either irradiated or non-irradiated produce. The investigation involved the actual sale of irradiated and non-irradiated apples to consumers. Limited information about the process was provided, and apples were sold at roadside stands. Prices for the irradiated apples were varied while the price for the non-irradiated apples was held constant. Of these 228 West-Central Missouri shoppers, 101 (44%) bought no irradiated apples, 86 (38%) bought only irradiated apples, and 41 (18%) bought some of both types, Results of probit regressions indicated three significant independent variables. There was an inverse relationship between the price of irradiated apples and the probability of purchasing irradiated apples. There was a positive relationship between the purchasers’ educational level and the probability of purchasing irradiated apples. Predicted probabilities for belonging to categories in probit models were computed. Depending on particular equation specification, correctly placed were approximately 70 percent of the purchasers of the two categories--bought only non-irradiated apples, or bought some of both irradiated and non-irradiated apples or only irradiated apples. Chronic ingestion of apple pectin has been shown to increase the absorption of quercetin in rats. The present study was designed to elucidate whether the simultaneous ingestion of quercetin with apple pectin could enhance the absorption of quercetin in humans, and the effects of dose dependency and degree of pectin methylation on quercetin absorption were also investigated. Healthy volunteers (n 19) received 200 ml of 0.5 mg/ml of quercetin drinks with or without 10 mg/ml of pectin each in a randomised cross-over design study with over 1-week intervals; urine samples from all the subjects were collected within 24 h after ingestion of the test drinks, and urinary deconjugated quercetin and its metabolites were determined using HPLC. The sum of urinary quercetin and its metabolites excreted was increased by 2.5-fold by the simultaneous ingestion of pectin. The metabolism of methylated quercetin (isorhamnetin and tamarixetin) was not affected by pectin ingestion. In six volunteers, who received quercetin drinks containing 0, 3 and 10 mg/ml of pectin, the sum of urinary quercetin and its metabolites excreted also increased in a pectin dose-dependent manner. Furthermore, the simultaneous ingestion of quercetin with low-methoxy and high-methoxy pectin, respectively, increased the sum of urinary excretion of quercetin and its metabolites by 1.69-fold and significantly by 2.13-fold compared with the ingestion of quercetin without pectin. These results elucidated that apple pectin immediately enhanced quercetin absorption in human subjects, and that its enhancing effect was dependent on the dose and degree of pectin methylation. The results also suggested that the viscosity of pectin may play a role in the enhancement of quercetin absorption. PMID:25865751

Full Text Available BACKGROUND: Human APPL1 and APPL2 are homologous RAB5 effectors whose binding partners include a diverse set of transmembrane receptors, signaling proteins, and phosphoinositides. APPL proteins associate dynamically with endosomal membranes and are proposed to function in endosome-mediated signaling pathways linking the cell surface to the cell nucleus. APPL proteins contain an N-terminal Bin/Amphiphysin/Rvs (BAR domain, a central pleckstrin homology (PH domain, and a C-terminal phosphotyrosine binding (PTB domain. Previous structural and biochemical studies have shown that the APPL BAR domains mediate homotypic and heterotypic APPL-APPL interactions and that the APPL1 BAR domain forms crescent-shaped dimers. Although previous studies have shown that APPL minimal BAR domains associate with curved cell membranes, direct interaction between APPL BAR domains on cell membranes in vivo has not been reported. METHODOLOGY: Herein, we used a laser-scanning confocal microscope equipped with a spectral detector to carry out fluorescence resonance energy transfer (FRET experiments with cyan fluorescent protein/yellow fluorescent protein (CFP/YFP FRET donor/acceptor pairs to examine interactions between APPL minimal BAR domains at the subcellular level. This comprehensive approach enabled us to evaluate FRET levels in a single cell using three methods: sensitized emission, standard acceptor photobleaching, and sequential acceptor photobleaching. We also analyzed emission spectra to address an outstanding controversy regarding the use of CFP donor/YFP acceptor pairs in FRET acceptor photobleaching experiments, based on reports that photobleaching of YFP converts it into a CFP-like species. Mixed life stages of obscure mealybug, Pseudococcus viburni (Signoret), and late 1st-instar or early 2nd-instar lightbrown apple moth, Epiphyas postvittana (Walker), on 'Royal Gala' apples (Malus domestica Borkhausen) were exposed to standard packhouse processing with and without addition of high-pressure apple washer treatments. Insect removal and mortality were assessed. After standard packhouse processing approximately 60% of P. viburni remained on their host apples. The 2 high-pressure apple washer treatments (500 and 800 psi at 2.0 rods/s) were equally effective and significantly reduced the number of P. viburni on apples compared with the packhouse control. High-pressure apple washer removal by location decreased in the following order: calyx cavity outside the sepals > cheek approximately stem cavity > or = calyx beneath sepals. About half of the E. postvittana larvae infesting apples was removed by standard packhouse processing. Removal rates were similar for all locations on open-calyxed apples. However, no removal occurred from the calyx beneath the sepals if the apple calyx was closed. All 4 high-pressure apple washer treatments tested (500 and 800 psi at 1.0 and 2.0 rods/s) halved the number of larvae on the apple exterior relative to the packhouse control. The pattern of removal for larvae on open-calyxed apples was calyx outside sepals approximately stem cavity > calyx beneath the sepals approximately cheek. A similar pattern was evident for larvae on closed-calyxed apples, except insects beneath the sepals evaded removal. The persistence of insects on the apple cheek reflects the high proportion of larvae inside tunnels in this location compared with other apple locations. China is in a dominant position in apple production globaly with both the largest apple growing area and the largest export of fresh apple fruits. However, the annual productivity of China’s apple is signiifcantly lower than that of other dominant apple producing countries. In addition, apple production is based on excessive application of chemical fertilizers and the nutrient use efifciency (especialy nitrogen) is therefore low and the nutrient emissions to the environment are high. Apple production in China is considerably contributes to farmers’ incomes and is important as export product. There is an urgent need to enhance apple productivity and improve nutrient use efifciencies in intensive apple production systems in the country. These can be attained by improved understanding of production potential, yield gaps, nutrient use and best management in apple orchards. To the end, priorities in research on apple production systems and required political support are described which may lead to more sustainable and environmental-friendly intensiifcation of apple production in China.

The compositional characteristics of untreated pure juice prepared from 88 apple varieties grown in the region of Eferding/Upper Austria were determined. Many of the analyzed varieties are noncommercial, old varieties not present in the market. The aim of the study was to quantitate the mineral, phosphate, trace elements, and polyphenolic content in order to identify varieties that are of particular interest for a wider distribution. Great variations among the investigated varieties could be found. This holds especially true for the total polyphenolic content (TPC) ranging from 103.2 to 2,275.6 mg/L. A clear dependence of the antioxidant capacity on the TPC levels was detected. Bioinformatics was employed to find specific interrelationships, such as Mg²⁺/Mn²⁺ and PO₄³⁻/K⁺, between the analyzed bio- and phytochemical parameters. Furthermore, special attention was drawn on putative effects of grafting on the phytochemical composition of apple varieties. By grafting 27 different apple varieties on two trees grown close to each other, it could be shown that the apple fruits remain their characteristic phytochemical composition. Finally, apple juice prepared from selected varieties was further characterized by additional biochemical analysis including cytotoxicity, epidermal growth factor receptor (EGFR) inhibition, and α-amylase activity tests. Cytotoxicity and inhibition of EGFR activation were found to be dependent on the TPC, while α-amylase activity was reduced by the apple juices independent of the presence of polyphenolic substances. Taken together selected apple varieties investigated within this study might serve as preferable sources for the development of apple-based food with a strong focus on health beneficial effects. Many natural products exhibit anti-inflammatory activity by suppressing excessive nitric oxide (NO) production by inducible NO synthase (iNOS). The maritime pine bark extract Pycnogenol has been formerly shown to decrease nitrite generation, taken as an index for NO, but so far it was not clear which constituent of the complex flavonoid mixture mediated this effect. The purpose of this study was to elucidate whether the in vivo generated Pycnogenol metabolite M1 (δ-(3,4-dihydroxyphenyl)-γ-valerolactone) displayed any activity in the context of induction of iNOS expression and excessive NO production. For the first time we show that M1 inhibited nitrite production (IC(50) 1.3 μg/ml, 95% CI 0.96-1.70) and iNOS expression (IC(50) 3.8 μg/ml, 95% CI 0.99-14.35) in a concentration-dependent fashion. This exemplifies bioactivation by metabolism because the M1 precursor molecule catechin is only weakly active. However, these effects required application of M1 in the low-micromolar range, which was not consistent with concentrations previously detected in human plasma samples after ingestion of maritime pine bark extract. Thus, we investigated a possible accumulation of M1 in cells and indeed observed high-capacity binding of this flavonoid metabolite to macrophages, monocytes, and endothelial cells. This binding was distinctly decreased in the presence of the influx inhibitor phloretin, suggesting the contribution of a facilitated M1 transport into cells. In fact, intracellular accumulation of M1 could explain why in vivo bioactivity can be observed with nanomolar plasma concentrations that typically fail to exhibit measurable activity in vitro. In the present study, 22 apple (Malus domestica Borkh. cultivars were tested in the filed for their relative resistance/susceptibility to WAA. These apple cultivars were found to respond differentially to WAA infestation. Based on tree infestation rating, cultivars were ranked into 6 relative resistance/susceptibility groups, as follows: immune - ‘Golden Delicious’, ‘Delbarestivale’, ‘Golden Smoothie’, ‘Red Miracle’ and ‘Harmony’; resistant - ‘Stark Gold’, ‘Early Gold’ and ‘Argi Gala’; moderately resistant - ‘Evanee’ and ‘Black Stayman’; moderately susceptible - ‘Vista Bella’, ‘Jonagold’, ‘Royal Gala’, ‘Jersey Mac’, ‘Granny Spur Type’ and ‘Summerred’; susceptible - ‘Jonathan’, ‘Nagava 6’, ‘Florina’, ‘Red Chief’ and ‘Gold Iralis’; highly susceptible - ‘Fuji 6’. Terpenes are specialized plant metabolites that act as attractants to pollinators and as defensive compounds against pathogens and herbivores, but they also play an important role in determining the quality of horticultural food products. We show that the genome of cultivated apple (Malus domestica) contains 55 putative terpene synthase (TPS) genes, of which only 10 are predicted to be functional. This low number of predicted functional TPS genes compared with other plant species was supported by the identification of only eight potentially functional TPS enzymes in apple 'Royal Gala' expressed sequence tag databases, including the previously characterized apple (E,E)-α-farnesene synthase. In planta functional characterization of these TPS enzymes showed that they could account for the majority of terpene volatiles produced in cv Royal Gala, including the sesquiterpenes germacrene-D and (E)-β-caryophyllene, the monoterpenes linalool and α-pinene, and the homoterpene (E)-4,8-dimethyl-1,3,7-nonatriene. Relative expression analysis of the TPS genes indicated that floral and vegetative tissues were the primary sites of terpene production in cv Royal Gala. However, production of cv Royal Gala floral-specific terpenes and TPS genes was observed in the fruit of some heritage apple cultivars. Our results suggest that the apple TPS gene family has been shaped by a combination of ancestral and more recent genome-wide duplication events. The relatively small number of functional enzymes suggests that the remaining terpenes produced in floral and vegetative and fruit tissues are maintained under a positive selective pressure, while the small number of terpenes found in the fruit of modern cultivars may be related to commercial breeding strategies. In Part 1, we reported the thermal distribution characteristics and the identification methods of apples, leaves and branches by using the infrared thermal image at the specific time. This paper reports the temperature changing characteristics and the relationships among apples, leaves and air temperature based on the information measured by the infrared thermal image equipment in the real-time for 24 hours. As a result, it was confirmed that the average temperature of apples was 1 degree C or more higher than the one of the leaves, and the average temperature of the leaves was almost same as the air temperature within daytime and about 3 hours period after sunset. It was also clarified for a remarkable temperature difference not to exist for midnight and the early morning between the apples and the leaves, and both became almost as well as the air temperature. After X-radiation of grafts of antonovka apple trees, three groups of morphological mutants, namely, weak-, average- and violently-growing, have been revealed. Although the mutation spectrum has some indefinite character a dose of 6 kR causes, more frequently and in a greater number, the weak-growing mutants, and a dose of 2 kR, the violently-growing ones. The advent of the 'personal' microcomputer provides a new tool for the debugging, calibration and monitoring of small scale physics apparatus, e.g., a single detector being developed for a larger physics apparatus. With an appropriate interface these microcomputer systems provide a low cost (1/3 the cost of a comparable minicomputer system), convenient, dedicated, portable system which can be used in a fashion similar to that of portable oscilliscopes. Here we describe an interface between the Apple computer and CAMAC which is now being used to study the detector for a Cerenkov ring-imaging device. Full Text Available The aim of the study was to assess the effect of three weeks of storage on the chemical and rheological properties of apple juices obtained from Idared and Jonatan apples variety. Total antioxidant activity, levels of bio-active compound groups and the viscosity were measured to characterize the investigated juices. The method applied for the determination of ascorbic acid concentration was with 2, 6-diclorophenolindophenol. Total phenols (TP in apple juices were determined using the Folin-Ciocalteau method and antioxidant activity by the use of DPPH free radical method. The viscosity of apple juices was investigated by a rotational viscometer, Brookfield viscometer (Brookfield Engineering Inc., Model RV-DV I Prime with RV spindles. During three weeks of storage, different rates of all measured properties have been observed decreasing for both studied apples varieties juices. The juices from Jonatan apples have higher antioxidant activities that are correlated with the higher content in polyphenols and lower values of viscosity.
The mutlivariate analysis of four year records on several vegetative and productive traits of twenty-one apple mutants (3 of 'Jonathan', 3 of 'Ozark Gold', 14 of 'Mollie's Delicious', 1 of 'Neipling's Early Stayman)' induced by gamma radiations showed that observation of some traits of one-year-old shoots is the most efficient way to reveal compact growing apple mutants. In particular, basal cross-section area, total length and leaf area resulted the most appropriate parameters, while internode length together with conopy height and width are less appropriate. The most interesting mutants we found are: one of 'Mollie's Delicious for the best balance among tree and fruit traits and for high skin color; one of 'Neipling's Early Stayman' with an earlier and more extensively red colored apple than the original clone. Escalating production costs, heavy reliance on non-renewable resources, reduced biodiversity, water contamination, chemical residues in food, soil degradation and health risks to farm workers handling pesticides all bring into question the sustainability of conventional farming systems. It has been claimed, however, that organic farming systems are less efficient, pose greater health risks and produce half the yields of conventional farming systems. Nevertheless, organic farming became one of the fastest growing segments of US and European agriculture during the 1990s. Integrated farming, using a combination of organic and conventional techniques, has been successfully adopted on a wide scale in Europe. Here we report the sustainability of organic, conventional and integrated apple production systems in Washington State from 1994 to 1999. All three systems gave similar apple yields. The organic and integrated systems had higher soil quality and potentially lower negative environmental impact than the conventional system. When compared with the conventional and integrated systems, the organic system produced sweeter and less tart apples, higher profitability and greater energy efficiency. Our data indicate that the organic system ranked first in environmental and economic sustainability, the integrated system second and the conventional system last. PMID:11309616

Full Text Available Interspecific hybridizations represent one of the apple breeding methods by which a wide variability can be achieved, useful for creating new cultivars. The study of 2190 interspecific hybrids, obtained from 25 combinations among crab apple species (Malus coronaria, M. floribunda, M. niedzwetzkyana, M. zumi, M. prunifolia and different apple cultivars, points out a large variability of the F1 seedlings for several traits, with significant importance in apple breeding programs. The first year of fructification, as mean per hybrid combination, varied from 6 (M. zumi x ‘Jonathan’ to 9.3 years; the average hybrid’s age for fructification was 7.4 years. The size of fruits among families varied from 1.5 (‘Starkrimson’ x M. prunifolia to 4.0 (‘Reinette Baumann’ x M. zumi, the mean being settled at 2.8 (therefore below mark 3, meaning ‘small fruits’. The lowest infection rate both for apple scab and powdery mildew attack was noticed at hybrids from M. coronaria x ‘Reinette Baumann’. On the whole, the hybrids with genitors of ‘species x cultivars’ type have had an early fruiting and a better resistance to scab and powdery mildew, compared to the ones from ‘cultivars x species’ combinations. The large variability of the studied traits gave the possibility to identify offsprings with desirable characteristics on nine hybrid combinations. Among these, 53 elite plants were selected, with a strength of selection of 2.42%, therefore a relative low value, correlated with the peculiarities of the interspecific population and the selection criteria for dessert apple.

Full Text Available As apple-processing units are now in expansion in Brazil, industrial by-products like pomace play an important role in pectin manufacture. The objective of this article was to determine a pratical follow-up to the extraction of pectin from apple pomace and to characterize it in a laboratory, on a small scale, aiming at establishing the optimum conditions for acid extraction. The highest yields were obtained when apple pomace was dried and ground to obtain an apple flour to be used as raw material,  citric or nitric acids were used and  when the citric acid concentration was 6.2 g/100 ml and the time of reaction was 153 minutes. The apple variety in itself was not significant in pectin yield. The degree of esterification (DE = 68.84 % of the product obtained, as well as its physical looks, show the success of pectin extraction.No aproveitamento de resíduos das indústrias processadoras de maçã, em expansão no Brasil, a pectina alimentícia surge como importante subproduto. Os objetivos deste trabalho foram os seguintes: extração e caracterização da pectina de bagaço de maçã em bancada, em que as condições para maior rendimento de obtenção em relação ao tipo de ácido, ao tamanho das partículas e à variedade de maçã foram estabelecidas. Objetivou-se também a otimização da produção, levando-se em conta o tempo e a concentração do ácido cítrico. Foram considerados métodos de extração mais eficientes quanto ao rendimento:  acidificação com ácido cítrico ou ácido nítrico,  cominuição do bagaço, usando a farinha como matéria-prima, com partículas retidas no tamis de 106 µm,  concentração de ácido de 6,2 g% e tempo de extração de 153 minutos. A variedade da maçã não interferiu significativamente no rendimento. A alta metoxilação (DE= 68,84% das pectinas produzidas em bancada confirma o grau de maturidade das matérias-primas usadas nos experimentos.
Studies on '60'Co-gamma irradiation of Golden Delicious apple have been carried out. The results showed that the optimum irradiation dosage for stored apple was ranged from 0.3-0.5 k Gy, with this dosage, the contents of vitamin c and titratalbe acidity in these apples had no significant change compared with unirradiated apples. This study evaluates the use of vacuum impregnation (VI) for developing nutritionally fortified fresh cut apples (Royal Gala). Cut apples were immersed in diluted high fructose corn syrup (20% w/w or 50% w/w) containing calcium or zinc. A vacuum pressure of 50 mmHg was applied for 15 min following atmospheric pressure restoration for 30 min while samples remained in the VI solution. Nutraceutical content and physicochemical properties of the apples immediately after VI were determined. Storability of VI apples at 6 degrees C and 90% relative humidity was also studied based on the color and texture of apples. Results indicated that 15-20% of the Daily Reference Intake of calcium and above 40% of the Daily Reference Intake of zinc could be obtained in 200 g fresh cut apples. VI treatments in 20% w/w high fructose corn syrup solutions had little effects on the physicochemical properties of apples. Storage study showed that VI with zinc significantly improved color stability, and calcium enhanced the firmness of the apples. PMID:12907403

Full Text Available A method to detect Apple stem grooving virus (ASGV based on reverse transcription polymerase chain reaction (RT-PCR was developed using primers ASGV4F-ASGV4R targeting the viral replicase gene, followed by a sandwich hybridisation, in microtiter plates, for colorimetric detection of the PCR products. The RT-PCR was performed with the Titan™ RT-PCR system, using AMV and diluted crude extracts of apple (Malus domestica leaf or bark for the first strand synthesis and a mixture of Taq and PWO DNA polymerase for the PCR step. The RT-PCR products is hybridised with both a biotin-labelled capture probe linked to a streptavidin-coated microtiter plate and a digoxigenin (DIG-labelled detection probe. The complex was detected with an anti-DIG conjugate labelled with alkaline phosphatase. When purified ASGV was added to extracts of plant tissue, as little as 400 fg of the virus was detected with this method. The assay with ASGV4F-ASGV4R primers specifically detected the virus in ASGV-infected apple trees from different origins, whereas no signal was observed with amplification products obtained with primers targeting the coat protein region of the ASGV genome or with primers specific for Apple chlorotic leaf spot virus (ACLSV and Apple stem pitting virus (ASPV. The technique combines the power of PCR to increase the number of copies of the targeted gene, the specificity of DNA hybridization, and the ease of colorimetric detection and sample handling in microplates.

The Brazilian crops mangoes and apples has expanding in the last years. However, tropical fruits crops are susceptible to infection that harms the crop yield. The control of these diseases is made through fungicides such as prochloraz which possesses protecting eradicating action by controlling anthracnose in mangoes and scabies in apples. Agrochemicals are of great importance in the agriculture considering the of relationship cost/benefit. However they may cause a series of problems in the ecosystem, being the levels of agrochemicals residues in fruits one of these factors. The aim of this work was to evaluate the prochloraz levels in mangoes treated in both stages of pre and post harvest, and in apples treated in the pre harvest. In the treatment of the mangoes, the interaction, fungicide with the gamma radiation with dose of 1,0 kGy, was used to induce prochloraz degradation. Treated post harvest mangoes were stored, at 12 deg C during 21 days. The results showed that the prochloraz did not present reduction in the residual levels of the mangoes after 21 day storage, that is the safe period established by the Brazilian legislation on agrochemicals, in treated mangoes. The refrigerated storage (12 deg C) and the gamma radiation also did not contribute to the degradation of the fungicide in mangoes. In average the concentration of the fungicide prochloraz in the peels (bar X = 1,64 mug/g) was higher than in the mangoes pulp (bar x = 0,06 mug/g), which allows the consumption of this fruit, since the peel is always discarded. The degradation product, formed in peels of mangoes in fruits treated in the pre and post harvest was the metabolite BIS 44596; for mangoes treated in the pre-post harvest the metabolite formed was the BIS 445186. The irradiation treatment of fresh fruits and vegetables for phytosanitary purposes is a satisfactory alternative method to others like fumigation and cold and hot treatments. Its use is increasing in several countries, and at present its approval is under revision by the National Regulatory Authorities. To verify the control process, apart from irradiation and dosimetry certificates, National Authorities require complementary evidence to show the efficacy of this treatment, especially when the documentation is not clear. The irradiation of fresh fruits produces single and double fragmentation in the DNA molecule, which can be measured using the microgel electrophoresis of individual cell (comet assay). The purpose of this work was to evaluate if it is possible to identify the irradiated apples for phytosanitary purposes from the others that were not treated. The possibility to estimate the absorbed dose was also evaluated. The methodology was carried out on the cell suspension obtained from irradiated seed cells with incremental doses (100, 200 and 300 Gy). The irradiation treatment for phytosanitary purposes to avoid emergency of codling moth ( Cydia pomonella) is 200 Gy. These results show that with this methodology it can be determined if the apple was irradiated or not. Full Text Available After 40 years of intensive research, chemistry of marine natural products has become a mature field. Since 1995, there are signals of decreased interest in the search of new metabolites from traditional sources such as macroalgae and octocorals, and the number of annual reports on marine sponges stabilized. On the contrary, metabolites from microorganisms is a rapidly growing field, due, at least in part, to the suspicion that a number of metabolites obtained from algae and invertebrates may be produced by associated microorganisms. Studies are concerned with bacteria and fungi, isolated from seawater, sediments, algae, fish and mainly from marine invertebrates such as sponges, mollusks, tunicates, coelenterates and crustaceans. Although it is still to early to define tendencies, it may be stated that the metabolites from microorganisms are in most cases quite different from those produced by the invertebrate hosts. Nitrogenated metabolites predominate over acetate derivatives, and terpenes are uncommon. Among the latter, sesquiterpenes, diterpenes and carotenes have been isolated; among nitrogenated metabolites, amides, cyclic peptides and indole alkaloids predominate.

Full Text Available Granny Smith apple pomace was subjected to evaluation as valuable source of antioxidant and anticancer phytochemicals on the basis of its content in phenolic compounds, antioxidant and antiproliferative activity. The total cotent of phenolics, flavonoids and flavan-3-ols in apple pomace determined spectrophotometrically, was 7.02 mg/g, 0.51 mg/g and 8.80 mg/g. Major phenolics (phenolic acids, flavan-3-ols, flavonoids and dihydrochalcons in apple pomace were identified and quantified by HPLC. The antioxidant activity of apple pomace on stable 1,1-diphenyl-2-picrylhydrazyl (DPPH and reactive hydroxyl radicals, was investigated by electron spin resonance (ESR spectroscopy. The IC50 DPPH and IC50 OH values of Granny Smith apple pomace were 9.51 mg/ml and 29.17 mg/ml, respectively. The antiproliferative activities of apple pomace on cervix epitheloid carcinoma (HeLa, colon adenocarcinoma (HT-29 and breast adenocarcinoma (MCF7 cell lines were determined according to the MTT (3-(4,5-dimethylthiazol-2-yl- 2,5-diphenyltetrazolium bromide colorimetric assay. The IC50 HeLa , IC50 HT-29 and IC50 MCF7 values of Granny Smith apple pomace were 26.40 mg/ml, 22.47 mg/ml and 21.26 mg/ml, respectively. Bramley apples were treated with Ridomil mbc 60 WP, containing carbendazim and metalaxyl, at half and full rate application at 10 and 20 degrees C, and with diphenylamine to control disease during storage. Carbendazim and metalaxyl residues were determined by HPLC and GC/MS, respectively, in apple core, flesh and peel at the initial time, 3 and 6 months after storage under controlled atmosphere conditions of 4.5 degrees C, 5% CO(2) and 1% O(2). The concentration of carbendazim residues in apple flesh was metalaxyl residues in apple flesh was metalaxyl in apple was 1.0 mg kg(-1). The temperature of the fungicide solution had little effect on the carbendazim residues but did have some effect on metalaxyl residues. Carbendazim residue content per apple was decreased when diphenylamine was included. A change in the ratio of carbendazim to metalaxyl was noted in the flesh, core and peel of the stored apple. The ratio of carbendazim to metalaxyl was 5:1 in the fungicide mix as applied. The ratio remained fairly constant in the core during storage. However, the ratio changed significantly in the peel and, to a lesser extent, and in a different direction, in the flesh. In the peel, the ratio was around 15:1 at 3 months of storage and 12:1 after 6 months of storage, whereas for flesh the ratios were 2:1 and 4:1, respectively. PMID:13129789
During the period 1973 - 79 studies were conducted with the aim of developing compact mutants in apple and cherry cultivars and in apple vegetative rootstocks. During the investigations the effect of the dose of gamma rays on frequency of the mutants was studied. Attempts were also made to evolve a micropropagation technique adapted to propagate P 2 and P 22 apple rootstocks, as an aid in mutation breeding. Several mutants were produced in all the material studied, but none of them have yet reached a sufficient developmental stage to enable their complete assessment. On the basis of the results obtained so far the following conclusions can be drawn: higher doses of irradiation resulted in higher frequency of mutants in most apple cultivars and apple rootstocks; in sour cherries the effect of dose depended on the cultivars. Among V1 shoots developed from sleeping buds on irradiated scion wood, compact mutants were found; their frequency, however, was about 60% lower than among V1 shoots developed directly from irradiated dormant buds. In apple rootstocks A 2 and M 26 several dwarfed mutants were found; some of these produced thorny plants and some had lower rooting ability; both these characteristics are inferior from the practical point of view. Multiplication and rooting media for in vitro propagation of apple rootstocks, worked out for M 26, were found unsuitable for the rootstocks P 2 and P 22; modifications made in the growth substance composition of the above media enabled satisfactory propagation to be obtained. People are regularly bombarded with logos in an attempt to improve brand recognition, and logos are often designed with the central purpose of memorability. The ubiquitous Apple logo is a simple design and is often referred to as one of the most recognizable logos in the world. The present study examined recall and recognition for this simple and pervasive logo and to what degree metamemory (confidence judgements) match memory performance. Participants showed surprisingly poor memory for the details of the logo as measured through recall (drawings) and forced-choice recognition. Only 1 participant out of 85 correctly recalled the Apple logo, and fewer than half of all participants correctly identified the logo. Importantly, participants indicated higher levels of confidence for both recall and recognition, and this overconfidence was reduced if participants made the judgements after, rather than before, drawing the logo. The general findings did not differ between Apple and PC users. The results provide novel support for theories of attentional saturation, inattentional amnesia, and reconstructive memory; additionally they show how an availability heuristic can lead to overconfidence in memory for logos. This study was conducted to analyze the ultrasonic transmitted signal for apple using wavelet transform. Fruit consists of nonlinear visco-elastic properties such as flesh, an ovary and rind and lienee most ultrasonic wave is attenuated and its frequency is shifted during passing the fruit. Thus it is not easy to evaluate the internal quality of the fruit using typical ultrasonic parameters such as wave velocity, attenuation, and frequency spectrum. A reversed-phase HPLC method using ion-pair formation has been developed for the simultaneous determination of naltrexone and three urinary metabolites. The extraction of the free and conjugated metabolites was studied by liquid-solid procedures using styrene-divinylbenzene copolymers (Amberlite XAD-2) and bonded octadecyl silica supports (ODS-silica). Optimum recovery was obtained with ODS-silica extraction using 25% acetonitrile in a 5 mM diammonium phosphate buffer pH 2.1 as elution solvent. The chromatographic behaviour of naltrexone metabolites and naloxone (internal standard) was examined by varying the mobile phase composition. Increments of both the diammonium phosphate buffer concentration and the percentage of organic solvent in the eluent decreases the retention of compounds in a non-linear manner. Increments of the dodecyl sulphate (counter-ion) concentration, increases the retention time. The method was applied to determine the urinary levels of major naltrexone metabolites in a volunteer receiving a 50 mg oral dose. This is the first method reported which permits the simultaneous quantitative determination of naltrexone and its metabolites, 6beta-naltrexol, naltrexone glucuronide and 6beta-naltrexol glucuronide, in urine. In the present study, to improve the therapeutic efficacy of phytodrugs and enhance photoprotective capability, we used poly(D,L-lactide-co-glycolide as a carrier of apple peel ethanolic extract (APETE on permeation-enhanced nanoparticles (nano-APETE. The in vitro toxicity of nano-APETE-treated dermal fibroblast cells were studied in a bioimpedance system, and the results coincided with the viability assay. In addition, the continuous real-time evaluations of photodamage and photoprotective effect of nano-APETE on cells were studied. Among three different preparations of nano-APETE, the lowest concentration provided small, spherical, monodispersed, uniform particles which show high encapsulation, enhanced uptake, effective scavenging, and sustained intracellular delivery. Also, the nano-APETE is more flexible, allowing it to permeate through skin lipid membrane and release the drug in a sustained manner, thus confirming its ability as a sustained transdermal delivery. In summary, 50 µM nano-APETE shows strong synergistic photoprotective effects, thus demonstrating its higher activity on target sites for the treatment of skin damage, and would be of broad interest in the field of skin therapeutics. Full Text Available Japanese ginjo sake is a premium refined sake characterized by a pleasant fruity apple-like flavor and a sophisticated taste. Because of technical difficulties inherent in brewing ginjo sake, off-flavors sometimes occur. However, the metabolites responsible for off-flavors as well as those present or absent in higher quality ginjo sake remain uncertain. Here, the relationship between 202 polar chemical compounds in sake identified using capillary electrophoresis coupled with time-of-flight mass spectrometry and its organoleptic properties, such as quality and off-flavor, was examined. First, we found that some off-flavored sakes contained higher total amounts of metabolites than other sake samples. The results also identified that levels of 2-oxoglutaric acid and fumaric acid, metabolites in the tricarboxylic acid cycle, were highly but oppositely correlated with ginjo sake quality. Similarly, pyridoxine and pyridoxamine, co-enzymes for amino transferase, were also highly but oppositely correlated with ginjo sake quality. Additionally, pyruvic acid levels were associated with good quality as well. Compounds involved in the methionine salvage cycle, oxidative glutathione derivatives, and amino acid catabolites were correlated with low quality. Among off-flavors, an inharmonious bitter taste appeared attributable to polyamines. Furthermore, protein analysis displayed that a diversity of protein components and yeast protein (triosephosphate isomerase, TPI leakage was linked to the overall metabolite intensity in ginjo sake. This research provides insight into the relationship between sake components and organoleptic properties.

In a study, the firmness of pear fruit during 4 weeks of storage was monitored using non-destructive impulse response (I-R and destructive Magness-Taylor (M-T puncture tests. The results of this study showed that the dominant frequency, stiffness coefficient and elasticity coefficient as a function of time could be expressed as a decreasing linear function (Gómez et al., 2005. Tiplica et al., (2010, showed that acoustic measurement can be a useful tool to discriminate different apple batches with a low error rate. Starting from the spectrum of the signal recorded by a microphone after the impact of a small hammer on the fruit, 18 key features were identified and used for the classification of apples belonging to 10 different varieties. The study aimed to evaluate apple firmness measured using both the penetrometer and acoustic methods. The methodologies were applied to Royal Gaya and Golden Smoothee apples harvested from 12 different orchards in Catalonia (Spain, on six different dates, and over three seasons. Chitosan is a good flocculant. The paper deals with the clarification of apple juice by means of chitosan. The results showed that the transmittance was over 97% and soluble solid content was stable basically, under the technological condition of chitosan of 0.5 - 1.2 g L-1 , temperature of 45 - 55℃ and pH 4.5. After the orthogonal trial, the optimum technological conditions of apple juice clarification by using chitosan were 0.3 g L-1 chitosan, 45℃C and pH 4.5. The research of the apple juice clarification with chitosan made a basis of the application of chitosan and provided the theoretical basis for the clarification of apple juice with chitosan.
Full Text Available Italy is one of the main apple producers in Europe, primarily intended for fresh consumption, both in the domestic and foreign markets. Fruit yield and quality depends on the cultivar, rootstock, and management practices, such as the fertilization and irrigation adopted in the orchard. This review aims at reporting the main apple cultivars and rootstocks, the management of fertilization and irrigation, as well as their adaptation to apple tree orchards in Italy. The programs for genetic improvement carried out in this country involved the selection of apple tree cultivars and rootstocks which enable a high fruit yield and quality, in order to meet the requirements from the consumer market. In the fertilization and irrigation management, nutrients and water are supplied in amounts next to the actual need of the plants, providing an adequate nutrition, a satisfactory yield, and high quality fruits, besides preventing, whenever possible, nutrients and water losses in the environment.
The activities of seven kinds of apples produced in Aomori prefecture were measured with a high pure Ge detector set in a 15 cm-thick lead shield. The activities of 40K were detected in all samples, but an artificial radioactive nuclide was not detected. The slices of an apple were contacted to an imaging plate for 15 days in a 15 cm-thick lead shield and cross sectional activity distributions were measured. By using the distribution map, the activities of skin, flesh and lead portion of an apple 'Ohrin' were calculated to 0.041, 0.032 and 0.046 Bq/g, respectively. This measuring system will be used for the activity estimations of the farm products, specially apples, because of that many atomic institutions are in operation and in the planning stage on Aomori prefecture. Full Text Available In recent years, epidemiological and biochemical studies have shown that eating apples is associated with reduction of occurrence of cancer, degenerative, and cardiovascular diseases. This association is often attributed to the presence of antioxidants such as ascorbic acid (vitamin C and polyphenols. The substances that hinder the presence of free radicals are also able to protect cells from aging. In our laboratory we used yeast, a unicellular eukaryotic organism, to determine in vivo efficacy of entire apples and their components, such as flesh, skin and polyphenolic fraction, to influence aging and oxidative stress. Our results indicate that all the apple components increase lifespan, with the best result given by the whole fruit, indicating a cooperative role of all apple components.

It's shown that application of dynamics method positively influences on the pectin yields and its main parameters. The exposure of fruit surfaces to direct sunlight during the summer months can result in sunburn damage. Losses due to sunburn damage are a major economic problem when marketing fresh apples. The objective of this study was to develop and validate a model for simulating fruit surface temperature (FST) dynamics based on energy balance and measured weather data. A series of weather data (air temperature, humidity, solar radiation, and wind speed) was recorded for seven hours between 11:00-18:00 for two months at fifteen minute intervals. To validate the model, the FSTs of "Fuji" apples were monitored using an infrared camera in a natural orchard environment. The FST dynamics were measured using a series of thermal images. For the apples that were completely exposed to the sun, the RMSE of the model for estimating FST was less than 2.0 °C. A sensitivity analysis of the emissivity of the apple surface and the conductance of the fruit surface to water vapour showed that accurate estimations of the apple surface emissivity were important for the model. The validation results showed that the model was capable of accurately describing the thermal performances of apples under different solar radiation intensities. Thus, this model could be used to more accurately estimate the FST relative to estimates that only consider the air temperature.
