In this work, we report for the first time on the analysis of genetic diversity within a set of 36 Tunisian Opuntia ficus indica (L.) Mill. ecotypes using RAPD markers.Random decamer primers were screened to assess their ability to detect polymorphisms in this plant crop. A colony PCR technique was applied for both genomic and chloroplast DNA in the green microalgae Chlorella. Of five different lysis buffers, Chelex-100 was superior for DNA extraction, PCR and DNA storage. It also was insensitive to variations in cell density. The conditions established for an improved PCR formulation are applicable for screening of genetically-engineered transformants as well as bioprospecting of natural microalgal isolates. Besides multiple Chlorella species, we also demonstrate the efficacy of Chelex-100 for colony PCR with a number of other microalgal strains, including Chlamydomonas reinhardtii, Dunaliella salina, Nannochloropsis sp., Coccomyxa sp., and Thalassiosira pseudonana. Access for independents to retail gas markets was a central concern in European policy reform efforts in the 1990s. Upstream oil and gas companies reacted with strategic intentions of forward integration. By late 2004, forward integration was still weak, however. An important explanation of the gap between announced strategic re-orientation and actual strategy implementation lies in the political failure of EU member states to dismantle market barriers to entry for independents. Measures of dispositional and transactional coping strategies, anxiety and alexithymia were filled out by 77 participants in a longitudinal study design. Statistical analyses were performed using general linear models and partial least squares path modelling, low-constraint methods that are particularly useful in the behavioural sciences. For the risk estimate of fatal malignancies, an effective dose was proposed on the basis of the assumption that the risk should be equal whether the whole body irradiated uniformly or whether there is non-uniform irradiation. The effective dose was defined by the product of organ or tissue doses and a weighting factor representing the proportion of risk factor for a fatal malignancy resulting from organ or tissue irradiation to the total malignant factor. The risk of malignancies can be derived by multiplying the malignant significant factor by the product of the risk factor and the effective dose. For the genetic risk, a significant factor was a relative child expectancy and organ or tissue doses were gonad doses. However all studies since (and including) Thompson, et al. 1989, emphasize the considerable geographic variation in genetic oil types and are in generalized agreement as to the genetic oil types present and their spatial and stratigraphic relationships. In particular it is accepted that the Gulf rim (both north and south) is encircled by a ring of Mesozoic carbonate-sourced oils and that there is also at least one major deep water trend of carbonate-sourced oils in the northern Gulf. It is also generally agreed that many of the Gulf rim oils are of Oxfordian-Kimmeridgian age, though some, like the Sunniland oils of Florida, are of lower Cretaceous origin. For the purposes of energy forests, biological pest control should be interpreted as any method of using natural organisms or their products for the regulation of herbivores, below the economic threshold. The organisms include the energy forest crop species and natural enemies of pest herbivores. Examples are provided of three-trophic-level interactions, including some on willows and poplars. Bottom-up effects may be particularly strong in juvenile Salicaceae, which are used in energy forest production, because plants are growing vigorously, and many herbivores respond positively. Some major pests on willows and poplars appear to have weak top-down regulation, and they may be influenced most by bottom-up effects such as plant resistance through genetic variation, ontogenetic aging and physiological aging. Only the final shallow position of OGW can be defined by geochemical indices of maturity. We have exploited {open_quotes}progeny testing{close_quotes} to map quantitative trait loci (QTL) underlying the genetic variation of milk production in a selected dairy cattle population. A total of 1,518 sires, with progeny tests based on the milking performances of >150,000 daughters jointly, was genotyped for 159 autosomal microsatellites bracketing 1645 centimorgan or approximately two thirds of the bovine genome. Using a maximum likelihood multilocus linkage analysis accounting for variance heterogeneity of the phenotypes, we identified five chromosomes giving very strong evidence (LOD score {ge} 3) for the presence of a QTL controlling milk production: chromosomes 1, 6, 9, 10 and 20. These findings demonstrate that loci with considerable effects on milk production are still segregating in highly selected populations and pave the way toward marker-assisted selection in dairy cattle breeding. 44 refs., 4 figs., 3 tabs.

Permian coal and Triassic mudstone from the Ordos Basin were pyrolyzed in a closed system using a gold tube technique. Carbon and hydrogen isotopes of the gases generated from pyrolysis were compared to Mesozoic gases in the basin to interpret the origin, maturity and any mixing of gases. Maturation trends for thermogenic methane from both coal and lacustrine kerogens in our experiment were found to be independent of heating rate, allowing their use for determination of gas provenance. Gases from a tectonically stable area like the Shanbei slope are derived mainly from Yanchang lacustrine kerogen, and gases in tectonically active areas consist of mixtures of coal-derived gases and oil-associated methane from deeply buried formations, as well as oil-associated gases and biogenic gases from shallow depth. The thermal maturity of the C{sub 2} and C{sub 3} gases is estimated to cover an equivalent vitrinite reflectance range from 0.7% to 1.2% R{sub o}, whereas C{sub 1} gas exhibits a wide ...

Molecular genetic markers can be used to identify quantitative trait loci (QTL) for thermal resistance and this has allowed characterization of a major QTL for knockdown resistance to high temperature in Drosophila melanogaster. The QTL showed trade-off associations with cold resistance under laboratory conditions. However, assays of thermal tolerance conducted in the laboratory may not necessarily reflect performance at varying temperatures in the field. Here we tested if lines with different genotypes in this QTL show different thermal performance under high and low temperatures in the field using a release recapture assay. We found that lines carrying the QTL genotype for high thermal tolerance were significantly better at locating resources in the field releases under hot temperatures while the QTL line carrying the contrasting genotype were superior at cold temperatures. Aerodynamic streamtube models are based on the conservation-of-momentum principle in a quasi-steady flow, by equating the forces of the rotor blades to the change in streamwise momentum through the turbine. An analytical model considering a multi-streamtube system divided into two parts has been developed to determine the aerodynamic blade loads and rotor performance of the Darrieus wind turbine (CARDAA computer code). Carbon isotope discrimination (?(13) C) is considered a useful indicator for indirect selection of grain yield (GY) in cereals. Therefore, it is important to evaluate the genetic variation in ?(13) C and its relationship with GY. A doubled haploid (DH) population derived from a cross of two common wheat varieties, Hanxuan 10 (H10) and Lumai 14 (L14), was phenotyped for ?(13) C in the flag leaf, GY and yield associated traits in two trials contrasted by water availability, specifically, rain-fed and irrigated. The PICC was used as a conduit for venous access in a way that has not been described previously in the literature. This research validates a novel approach for source tracking based on denaturing gradient gel electrophoresis (DGGE) analysis of DNA extracted from Escherichia coli isolates. Escherichia coli from different animal sources and from river samples upstream from, at, and downstream of a combined sewer overflow were subjected to DGGE to determine sequence variations within the 16S-23S intergenic spacer region (ISR) of the rrnB ribosomal operon. The ISR was analyzed to determine if E. coli isolates from various animal sources could be differentiated from each other. DNA isolated from the E. coli animal sources was PCR amplified to isolate the rrnB operon. To prevent amplification of all 7 E. coli ribosomal operons by PCR amplification using universal primers, sequence-specific primers were utilized for the rrnB operon. Transient Enhanced Diffusion (TED) of dopants in Si is the consequence of the evolution, upon annealing, of a large supersaturation of Si self-interstitial atoms left after ion bombardment. In the case of amorphizing implants, this supersaturation is located just beneath the c/a interface and evolves through the nucleation and growth of End-Of-Range (EOR) defects. For this reason, the authors discuss here the relation between TED and EOR defects. Modelling of the behavior of these defects upon annealing allows one to understand why and how they affect dopant diffusion. This is possible through the development of the Ostwald ripening theory applied to extrinsic dislocation loops. Heterochromatin constitutes a significant portion of the genome in higher eukaryotes; approximately 30% in Drosophila and human. Heterochromatin contains a high repeat DNA content and a low density of protein-encoding genes. In contrast, euchromatin is composed mostly of unique sequences and contains the majority of single-copy genes. Genetic and cytological studies demonstrated that heterochromatin exhibits regulatory roles in chromosome organization, centromere function and telomere protection. As an epigenetically regulated structure, heterochromatin formation is not defined by any DNA sequence consensus. Heterochromatin is characterized by its association with nucleosomes containing methylated-lysine 9 of histone H3 (H3K9me), heterochromatin protein 1 (HP1) that binds H3K9me, and Su(var)3-9, which methylates H3K9 and binds HP1. Based on a literature study an actual summary of the risk-utility problems in X-ray diagnosis is outlined due to the Recommendations of the International Commission for Radiation Protection (ICRP publication 26/1977). Papers demonstrating quantitative assessment of the somatic and genetical risk in X-ray examination are preferably cited and evaluated. It is concluded that the somatic and genetical risk is low in diagnostic ratiology. However, it must not be neglected and has always to be compared to the utility of an examination as well as to other risks of the examination. (author).

We combine establishment-level employment data with trade data by port to examine the impact of accession to the European Economic Community on the spatial distribution of UK manufacturing. We test the predictions from economic geography models of how external trade affects the spatial distribution of employment. We treat accession as a natural experiment which helps mitigate problems of endogeneity. Our results suggest that accession changed the country composition of trade and via the port composition induced an exogenous shock to different locations. In line with theory, we find that better access to export markets and intermediate goods increase employment while increased import competition decreases employment.

The general development of variational methods for fusion reactor blanket studies is given. Important quantities such as tritium breeding ratio and total nuclear heating are linear functionals of the solutions to the Boltzmann transport equation. To estimate a neutronic quantity by variational methods is, in general, to carry out the scalar product formulation of the Roussopoulos variational principle, or the Schwinger variational principle, with the help of the associated adjoint transport equation where the appropriate response function for the estimate is taken as the source. We present numerical calculations for cesium and mercury to estimate the sensitivity of the scattering length to the variation of the fine structure constant alpha. The method used follows ideas Chin and Flambaum [Phys. Rev. Lett. 96, 230801 (2006)], where the sensitivity to the variation of the electron to proton mass ratio, beta, was considered. We demonstrate that for heavy systems, the sensitivity to variation of alpha is of the same order of magnitude as to variation of beta. Near narrow Feshbach resonances the enhancement of the sensitivity may exceed nine orders of magnitude.

The application of multi-objective genetic algorithms for green building design in two phases were presented in order to better help designers in the decision-making process. The purpose is to minimize two conflicting criteria: the life-cycle cost and the life-cycle environmental impact. Environmental impact criteria examined include energy and non-energy natural resources, global warming, and acidification. Variables focus on building envelope-related parameters. The application of multi-objective genetic algorithms is divided into two phases. The first phase intends to help designers in understanding the trade-off relationship between the two conflicting criteria. The second phase intends to refine the performance region that is of the designer's interest. In this paper, a new design methodology for determining the size, location, type and number of capacitors to be placed on a radial distribution system is presented. The objective is to minimize the peak power losses and the energy losses in the distribution system considering the capacitor cost. A sensitivity analysis based method is used to select the candidate locations for the capacitors. A new optimization method using a Genetic Algorithm is proposed to determine the optimal selection of capacitors. Test results have been presented along with the discussion of the algorithm.

The risks from radiation exposure during radiological diagnosis has usually been estimated in relation to genetic changes. Relevant information has been expressed as the genetically significant dose. In this paper we attempt to produce an analogous measure for evaluating the somatic risk in the form of a somatically significant dose index for radiological and CT examinations. It is shown that, for both types of examination, the two risk factors may be entirely different.

Abstract Polymorphisms in the genomic DNA of eight varieties maintained by conventional bud propagation (via rhizomes) and by in vitro shoot tip cultures were detected by RAPD analysis of sugarcane varieties. The study estimated the genetic diversity induced after in vitro multiplication of these varieties. Higher (28.9%) and lower (12%) numbers of polymorphic bands were detected in plants propagated via rhizomes; the genetic similarity estimate varying from 0.63 to 0.80. Calculating the local spectral densities at magnetic adatoms, we estimate the variation in the Yosida-Kondo resonance due to the Ruderman-Kittel-Kasuya-Yosida (RKKY) interaction and the direct exchange between adatoms in systems of magnetic trimers on metal surfaces. The results show that the RKKY interaction leads to the gradual variation in Kondo temperature, and the direct exchange can be the origin of the drastic variation. (author)

The translation of copies of this genetic blueprint (in the form of messenger RNA) into polypeptides is performed on the ribosome, a highly complex molecular machine composed of RNAs and proteins. To this end, special adaptor molecules called transfer RNAs are lined up by the ribosome in the sequence dictated by the genetic code, such that the amino acids carried by these molecules can be linked into a polypeptide. Several cofactors are involved in these processes, some of which require energy freed up by GTP hydrolysis. Genetic mapping is approached using the techniques of high resolution fluorescence in situ hybridization (FISH). This technology and the results of its application are designed to rapidly generate whole genome as tool box of expressed sequence to speed the identification of human disease genes. The results of this study are intended to dovetail with and to link the results of existing technologies for creating backbone YAC and genetic maps. In the first eight months, this approach generated 60--80% of the expressed sequence map, the remainder expected to be derived through more long-term, labor-intensive, regional chromosomal gene searches or sequencing. Three in vivo assays were used to measure the immunocompetence of chickens in two generations of a selection experiment. The obtained data were used to estimate the variance components for sire and dam for antibody production to Pasteurella multocida and Mycoplasma gallisepticum vaccines, for T-cell-mediated immunity evaluated by a phytohemagglutinin wing web assay, and for clearance of foreign particles from the circulatory system. Heritabilities of and genetic correlations among these immunological traits were calculated from the sire variance components. Heritability estimates of the immunological traits based on the sire component of variance ranged from .06 to .53, and genetic correlations among immunological traits were generally negative.
