Overexpression of a maize dehydrin gene, ZmDHN2b, in tobacco enhances tolerance to low temperature

British Library Electronic Table of Contents (United Kingdom)

Dehydrins, a subfamily of group 2 LEA proteins, are intrinsically unstructured plant proteins that accumulate in the late stages of seed development and in vegetative tissues subjected to water deficit, salinity, low temperature, or abscisic acid treatment. In this study, we isolated and characterized ZmDHN2b, a maize dehydrin gene. The genomic organization of the ZmDHN2b gene and its expression in maize seedlings were analyzed. To investigate the function of ZmDHN2b, we generated transgenic tobacco plants constitutively overexpressing ZmDHN2b. Ectopic expression of ZmDHN2b in tobacco accelerated seed germination and seedling growth at 15?C. Furthermore, ZmDHN2b-overexpressing lines had lower levels of cold-induced malondialdehyde and ...

Transport and superconducting properties of RNi_2B_2C (R=Y, Lu) single crystals

International Nuclear Information System (INIS)

The in-plane resistivity, in-plane absolute thermopower, and upper critical field measurements are reported for single-crystal samples of YNi_2B_2C and LuNi_2B_2C superconductors. The in-plane resistivity shows metallic behavior and varies approximately linearly with temperature near room temperature (RT) but shows nearly quadratic behavior in temperature at low temperatures. The YNi_2B_2C and LuNi_2B_2C single-crystal samples exhibit large transverse magnetoresistance (#approx#6 8% at 45 kOe) in the ab plane. The absolute thermopower S(T) is negative from RT to the superconducting transition temperature T_c. Its magnitude at RT is a few times of the value for a typical good metal. S(T) is approximately linear in temperature between #approx#150 K and RT. Extrapolation to T=0 gives large intercepts (few #mu#V/K) for both ...

Synthesis, structural activity-relationships, and biological evaluation of novel amide-based allosteric binding site antagonists in NR1A/NR2B N-methyl-d-aspartate receptors

British Library Electronic Table of Contents (United Kingdom)

The synthesis and structure-activity relationship analysis of a novel class of amide-based biaryl NR2B-selective NMDA receptor antagonists are presented. Some of the studied compounds are potent, selective, non-competitive, and voltage-independent antagonists of NR2B-containing NMDA receptors. Like the founding member of this class of antagonists (ifenprodil), several interesting compounds of the series bind to the amino terminal domain of the NR2B subunit to inhibit function. Analogue potency is modulated by linker length, flexibility, and hydrogen bonding opportunities. However, unlike previously described classes of NR2B-selective NMDA antagonists that exhibit off-target activity at a variety of monoamine receptors, the compounds described herein show much diminished effects against the...

Tissue-specific expression of Fgfr2b and Fgfr2c isoforms, Fgf10 and Fgf9 in the developing chick mandible

British Library Electronic Table of Contents (United Kingdom)

SummaryExperimental evidence has demonstrated the importance of FGF signalling in morphogenesis of the mandibular processes. FGFs transmit their signals through four tyrosine kinase transmembrane receptors (FGFRs). Alternative splicing in FGFRs including FGFR2 generates different isoforms that exhibit different ligand-specificities, exclusive tissue distributions and specific biological functions. Despite extensive information regarding the isoform-specific patterns of expression Fgfr2c and Fgfr2b during morphogenesis of many organs, a comparative analysis of these specific isoforms in the chick mandible has not been reported. To better understand the function of FGFR2 in mandibular morphogenesis, we have analysed the expression Fgfr2b, Fgfr2c and their putative ligands Fgf10 and Fgf9, in ...

Synthesis, characterization and evaluation of antibacterial activity of some thiazolo[3,2-b][1,2,4]triazole incorporating diphenylsulfone moieties

British Library Electronic Table of Contents (United Kingdom)

A series of thiazolo[3,2-b][1,2,4]triazole incorporating diphenylsulfone moieties were synthesized starting from 5-[4-(4-X-phenylsulfonyl)phenyl]-4H-1,2,4-triazole-3-thioles 3a-c, X=H, Cl, Br. Thus, alkylation of 1,2,4-triazoles 3 with phenacyl bromide or 4-bromophenacyl bromide afforded S-substituted 1,2,4-triazoles 4, 5. These new intermediates 4 and 5, in the presence of H2SO4 (c), were cyclized to 2-[4-(4-X-phenylsulfonyl)phenyl]-6-(4-Y-phenyl)[1,3]thiazolo[3,2-b]-[1, 2,4]-triazoles 6, 7 (I) and not to isomeric thiazolo[2,3-c][1,2,4]-triazoles 6, 7 (II). The newly synthesized compounds were characterized by IR, 1H, 13C NMR and elemental analysis. MS spectra confirmed the formation of thiazolo[3,2-b][1,2,4]triazole 6, 7 (forms I) in detriment of [2,3-c] isomeric compounds (forms II). Th...

Mott-Schottky analysis shows that for high enough electrode potential, and whatever the surface finish, the films behave like n-type semiconductors. 2B passive film appears to be more donor-doped than BA one and the density of donor states increases with chloride concentration. The electron donor levels are assumed to be generated by negatively charged cations vacancies produced by the chloride ions reaction with the outer passive film. Time-dependent infrared emission following photodissociation of nitromethane and chloropicrin.

Science.gov (United States)

Nitromethane (CH(3)NO(2)) and its chlorinated analogue, chloropicrin (CCl(3)NO(2)), were photolyzed at 193, 248, and 266 nm, and the products were observed by time-dependent Fourier transform infrared emission spectroscopy. At 193 and 248 nm, the primary photodissociation pathway for nitromethane was cleavage of the C-N bond to produce CH(3) + NO(2)(A (2)B(2)). At 266 nm, weak emission was observed following photodissociation of nitromethane, but an infrared spectrum could not be obtained. The photodissociation of chloropicrin at 193 nm produced the analogous product channel CCl(3) + NO(2)(A (2)B(2)) in addition to several other product channels. At 248 and 266 nm, only CCl(3) + NO(2)(A (2)B(2)) was observed. The production of phosgene (CCl(2)O) from chloropicrin photodissociation was not observed in this study. PMID:16571044

The details and principles of an apparatus built for measurements of fluorescence quantum yields and cascade-free lifetimes of open-shell cations are reported. These rely on the detection of coincidences between energy selected photo-electrons and undispersed photons. The results of such measurements for CO"+_2,COS"+,CS"+_2 and N_2O"+ in selected vibrational levels of their excited states are presented. Non-unity fluorescence quantum yields are found for some vibronic levels of CO"+_2(B), COS"+(A), N_2O"+(A) and a non-exponential decay is observed for CS"+_2(B). The data yield the following values for the radiative lifetimes: CO"+_2(A) 124 +- 6 ns,CO"+_2(B) 140 +- 7 ns, COS"+(A) 550 +- 50 ns and N_2O"+(A) 240 +- 12 ns. (orig.).

EPR, optical, infrared and Raman studies of VO"2"+ ions in polyvinylalcohol films

International Nuclear Information System (INIS)

Electron paramagnetic resonance (EPR), optical, infrared and Raman spectral studies have been carried out on vanadyl ions doped in polyvinylalcohol (PVA) films. The spin-Hamiltonian parameters (g and A) and the molecular orbital coefficients (#beta#_2"*"2 and k) have been evaluated. The values of spin-Hamiltonian parameters confirm that the vanadyl ions are present in PVA films as VO"2"+ molecular ions in an octahedral site with a tetragonal compression (C_4_v). The temperature variation EPR studies reveal that the variation of number of spins with temperature is in accordance with Boltzmann law. It is interesting to observe that the variation of susceptibility with temperature obeys Curie-Weiss law. The FT-IR and FT-Raman spectrum exhibits few bands, which are attributed to O-H, C-H, C-C and C-O groups of stretching and bending vibrations. Levels of mesenchymal FGFR2 signaling modulate smooth muscle progenitor cell commitment in the lung

British Library Electronic Table of Contents (United Kingdom)

Fibroblast growth factor (FGF) signaling has been shown to regulate lung epithelial development but its influence on mesenchymal differentiation has been poorly investigated. To study the role of mesenchymal FGF signaling in the differentiation of the mesenchyme and its impact on epithelial morphogenesis, we took advantage of Fgfr2c+/? mice, which due to a splicing switch express Fgfr2b in mesenchymal tissues and manifest Apert syndrome-like phenotypes. Using a set of in vivo and in vitro studies, we show that an autocrine FGF10?FGFR2b signaling loop is established in the mutant lung mesenchyme, which has several consequences. It prevents the entry of the smooth muscle progenitors into the smooth muscle cell (SMC) lineage and results in reduced fibronectin and elastin deposition. Levels of...

Immunosuppressant calcineurin inhibitors phase shift circadian rhythms and inhibit circadian responses to light

British Library Electronic Table of Contents (United Kingdom)

PP2B is a Ca2+/calmodulin-dependent protein phosphatase that is ubiquitously expressed in mammals. Among other actions, it is an effector mechanism in NMDA-mediated glutamate neurotransmission as well as a regulator of GSK3? and MAPK signaling cascades. Because all of these mechanisms have demonstrable roles in the control of circadian rhythyms, we hypothesized that PP2B would be a key regulator of rhythm generation and entrainment, and that through inhibition of its phosphatase activity, the circadian system would be affected by immunosuppressant drug therapy. We report here that immunosuppressant drugs (cyclosporin A, FK506) (1) block the circadian responses to light that underlie photic entrainment; (2) produce circadian phase shifts with a characteristic nonphotic profile; and (3) disr...

Structural instability of wild-type fibroblast growth factor (FGF)-1 and its dependence on exogenous heparin for optimal activity diminishes its potential utility as a therapeutic agent. Here we evaluated FGFC, an FGF1:FGF2 chimeric protein, for its receptor affinity, absolute heparin-dependence, stability and potential clinical applicability. Using BaF3 transfectants overexpressing each FGF receptor (FGFR) subtype, we found that, like FGF1, FGFC activates all of the FGFR subtypes (i.e., FGFR1c, FGFR1b, FGFR2c, FGFR2b, FGFR3c, FGFR3b and FGFR4) in the presence of heparin. Moreover, FGFC activates FGFRs even in the absence of heparin. FGFC stimulated keratinocytes proliferation much more strongly than FGF2, as would be expected from its ability to activate FGFR2b. Basic properties and battery performances of the novel high temperature stable lithium salt (Li{sub 2}B{sub 12}F{sub 12}, Dilithium Dodecafluorododecaborate; Li{sub 2}DFB) were studied using a Mn-based cathode and anode composed of a hard carbon and graphite mixture. The effect of co-solvents (mainly linear carbonate in electrolyte formulation of PC/EC/co-solvent (5/30/65 vol% mixture)) on conductivity, viscosity, charge-discharge capacities, rate performance, temperature performance, cycle life and storage life at 60 C was investigated. Conductivity of Li{sub 2}DFB electrolyte increased with reducing its viscosity by changing co-solvent and increasing the volume of the higher dielectric solvent. Li{sub 2}DFB electrolytes showed comparable discharge capacity and columbic efficiency against LiPF{sub 6} electrolyte. Agonist-directed trafficking of signalling at serotonin 5-HT2A, 5-HT2B and 5-HT2C-VSV receptors mediated Gq/11 activation and calcium mobilisation in CHO cells.

Science.gov (United States)

Several examples of agonist-directed trafficking of receptor signalling at 5-HT2A and 5-HT2C receptors have been reported that involve independent downstream transduction pathways. We now report the functional selectivity of a series of chemically diverse agonists at human (h)5-HT2A, h5-HT2B and h5-HT2C-VSV by examining two related responses, the upstream activation of Gq/11 proteins in comparison with its associated cascade of calcium mobilisation. At the h5-HT2A receptor, d-lysergic acid diethylamide (LSD) and the antiparkinsonian agents lisuride, bromocriptine and pergolide exhibit a higher potency for Gq/11 activation than calcium release in contrast with all the other tested ligands such as 5-HT, mCPP and BW723C86, that show an opposite preference of signalling pathway. Comparable observations are made at h5-HT2B and h5-HT2C-VSV receptors, suggesting a similar mechanism of functional selectivity ...

Zonal differences in DNA synthesis activity and cytochrome P450 gene expression in livers of male F344 rats treated with five nongenotoxic carcinogens

Energy Technology Data Exchange (ETDEWEB)

Both increased cell proliferation and {open_quotes}altered{close_quotes}CYP gene expression are prominent phenomena associated with liver tumor promotion by nongenotoxic carcinogen treatment. BRDU-labeled parenchymal nuclei were observed primarily in the periportal area of groups of rats, independent of nongenotoxic carcinogen treatment. Treatment with each of the 5 nongenotoxic carcinogens resulted in profound alterations in CPY gene expression at both the transcriptional and translational levels. Expression of CYP1A1, 1A1/2, 3A1, 2B1/2, and 4A immunoproteins demonstrated nongenotoxic carcinogen-specific patterns in both magnitude and zonal distribution. In agreement with the CYP immunoprotein data, treatment with each of the five nongenotoxic carcinogens resulted in a unique composition of mRNAs of CYP2B1, 2B2, 2C6, 2C11, 3A1, 3A2, and 4A1, which were variably ...

Simulation of dopant diffusion and activation during flash lamp annealing

International Nuclear Information System (INIS)

A set of advanced models implemented into the simulator Sentaurus Process was applied to simulate ultra shallow junction formation by flash lamp annealing (FLA). The full path transient enhanced diffusion model includes equations for small interstitial clusters (I_2, I_3, I_4), #left brace#3 1 1#right brace# defects and dislocation loops. A dopant-point defect clustering model is used for dopant activation simulation. Several cluster types are considered: B_2, B_2I, B_2I_2, B_3I, B_3I_2, B_3I_3 for boron and As_2, As_2V, As_3, As_3V, As_4, As_4V for arsenic. Different point defect and dopant-point defect pair charge states are taken into account to obtain accurate results in the high doping level region. The flux expressions in the three-phase segregation model include a dependence on the doping level and point defect supersaturation. The FLA process was ...

Texture transformations in thermomechanically treated steels having increased nitrogen content and alloys

International Nuclear Information System (INIS)

In this study the results of mathematical simulation and experimental research of texture transition in stress assisted shear phase transformations are discussed. The orientation distribution function (ODF) calculation model to the any type crystalline lattice symmetry materials has been suggested. By crystalline lattice symmetry analysis this computer program determines necessary amount of the initial experimental data. The experimental verification supports this model well. The ODF calculation model after shear phase transformation with equal use of all possible variants of orientation relationship or with various degrees of variant selection worked out. In this study the experimental and calculating data of the direct and reversal shear transformations #alpha# <=> #gamma# in Fe-alloys and B2 <=> B19' in Ti-Ni alloys are discussed. The stress assisted shear phase transformation model and computer program to calculate ODF after ...

Study on the crystallization behaviour and thermal stability of glass-ceramics used as solid oxide fuel cell-sealing materials

British Library Electronic Table of Contents (United Kingdom)

Glass ceramics are commonly used as sealing materials for planar solid oxide fuel cells (SOFCs). The major requirements of stack and module builders for these materials are the stability of the coefficient of thermal expansion (CTE), excellent bonding (sticking) behaviour and the absence of volatile ingredients, which can lead to changes of the material properties and the sealing ability. SCHOTT Electronic Packaging has developed special glasses and glass-ceramics for various solid oxide fuel cell designs and operating temperatures. The glass compositions are based on the system MgO-Al2O3-BaO-SiO2-B2O3. In this study the evaluation of the developed materials was done by high temperature aging tests for up to 1000h, high temperature XRD-studies and Rietveld calculations, combined with scann...

Specific features of the current-voltage characteristics of diffuse glow discharges in Ar:N2 mixtures

British Library Electronic Table of Contents (United Kingdom)

The characteristics of diffuse glow discharges in pure argon and the Ar + 1%N2 mixture at pressures of 2?80 Torr were studied experimentally and numerically. The discharge operated in a molybdenumglass tube with an inner diameter of 2.8 cm and interelectrode distance of 75 cm. The current-voltage characteristic of the discharge and the populations of the N2(B3? g ) and N2(C3? u ) states were measured. It is shown that, at relatively low pressures (P 15 Torr), the current-voltage characteristic of a discharge in the mixture lies lower than that in pure argon. As the pressure increases, the effect of the reduction in the discha...

Real-time neutron diffraction study of crystallization kinetics in amorphous Fe_7_8B_1_3Si_9 alloy

International Nuclear Information System (INIS)

The transformation from the as-quenched amorphous to the crystalline state in Fe_7_8B_1_3Si_9 alloy has been investigated using a new-type neutron diffractometer. The time resolved diffraction patterns clearly show that this alloy crystallizes into FeSi alloy and Fe_2B in that order. The time evolution of these crystalline phases can be analyzed by the Kolmogorov-Johnson-Mehl-Avrami equation with the exponent of about 2.5 over a wide temperature range. This suggests that the crystallization occurs by the diffusion-controlled growth with a constant nucleation rate. The scaling behavior in the crystallization kinetics is also discussed. (author).

Moessbauer investigation of the magnetic anisotropy and electronic structure of a metallic glass

International Nuclear Information System (INIS)

The temperature dependence of the average magnetization direction in an Fe_8_0B_2_0 glassy alloy (METGLAS 2605) is investigated by Moessbauer spectroscopy. Moessbauer spectra were recorded between 77 K and 363 K using a conventional constant-acceleration Moessbauer spectrometer with 10 mCi "5"7Co in Pd source. The direction of the easy magnetization is found to be strongly terperature dependent and the out-of-plane component turned to be unusually large. Comparing the magnetic moments some similarities between the electronic structure of the glassy alloy and the Fe_2B and FeB intermetallic compounds are established and discussed. (Sz.N.Z.).

Magnetism and recoilless fraction in metallic glasses after laser irradiation

British Library Electronic Table of Contents (United Kingdom)

Samples of Fe78B13Si9 (Metglas 2605 TCA) and Fe77Cr2B16Si5 (Metglas 2605 S3A) were exposed to pulsed excimer laser irradiation at repetition rates of 15, 25 and 50Hz. The occurrence of out-of-plane magnetic anisotropy, changes in the average hyperfine magnetic field and width of the hyperfine field distribution were evidenced by transmission Mossbauer spectroscopy. The variation of the recoilless fraction with laser processing parameters was derived from the simultaneous recording of Mossbauer spectra using a stainless steel etalon as a dual absorber. Conclusions on the onset of long-range magnetic order were formulated.

Crystal structure of the human centromeric nucleosome containing CENP-A

British Library Electronic Table of Contents (United Kingdom)

In eukaryotes, accurate chromosome segregation during mitosis and meiosis is coordinated by kinetochores, which are unique chromosomal sites for microtubule attachment. Centromeres specify the kinetochore formation sites on individual chromosomes, and are epigenetically marked by the assembly of nucleosomes containing the centromere-specific histone H3 variant, CENP-A. Although the underlying mechanism is unclear, centromere inheritance is probably dictated by the architecture of the centromeric nucleosome. Here we report the crystal structure of the human centromeric nucleosome containing CENP-A and its cognate ?-satellite DNA derivative (147 base pairs). In the human CENP-A nucleosome, the DNA is wrapped around the histone octamer, consisting of two each of histones H2A, H2B, H4 and CENP...

Correlated accumulation of anthocyanins and rosmarinic acid in mechanically stressed red cell suspensions of basil (Ocimum basilicum)

British Library Electronic Table of Contents (United Kingdom)

A red basil cell line (T2b) rich in rosmarinic acid (RA) was selected for the stable production of anthocyanins (ACs) in the dark. Cell suspension cultures were subjected to mechanical stress through increased agitation (switch from 90 to 150rpm) to determine the relationship between AC and RA accumulation. Cell extracts were analyzed by HPLC and LC-MS, and the resulting data were processed with multivariate statistical analysis. MS and MS/MS spectra facilitated the putative annotation of several complex cyanidin-based ACs, which were esterified with coumaric acid and, in some cases, also with malonic acid. It was also possible to identify various RA-related molecules, some caffeic and coumaric acid derivatives and some flavanones. Mechanical stress increased the total AC and RA contents, ...

Two subpopulations of stem cells for T cell lineage

Energy Technology Data Exchange (ETDEWEB)

An assay system for the stem cell that colonizes the thymus and differentiates into T cells was developed, and by using this assay system the existence of two subpopulations of stem cells for T cell lineage was clarified. Part-body-shielded and 900-R-irradiated C57BL/6 (H-2b, Thy-1.2) recipient mice, which do not require the transfer of pluripotent stem cells for their survival, were transferred with cells from B10 X Thy-1.1 (H-2b, Thy-1.1) donor mice. The reconstitution of the recipient's thymus lymphocytes was accomplished by stem cells in the donor cells and those spared in the shielded portion of the recipient that competitively colonize the thymus. Thus, the stem cell activity of donor cells can be evaluated by determining the proportion of donor-type (Thy-1.1+) cells in the recipient's thymus. Bone marrow cells were the most potent source of stem cells. By contrast, when the stem ...

Regulation of NMDA and AMPA receptors during the maturation phase of chicken brain development

International Nuclear Information System (INIS)

Full text: The maturation of chicken forebrain is protracted and occurs well after synapse formation providing a good model for studying mechanisms of brain maturation. Using microslices from immature (10 day) and adult chicken forebrain prepared after decapitation, we have examined functional properties of NMDA and AMPA receptors by measuring agonist-induced uptake of "4"5Ca"2"+ . The rate and extent of NMDA induced "4"5Ca"2"+ accumulation decreased during maturation with no change in EC_5_0. The rate and extent of the AMPA induced response also decreased with a 60-fold increase in EC_5_0. However, the total NMDA receptor content did not change as indicated by 3 H-MK801 binding and NR1 immunoreactivity in P2 fractions. Similarly, there was no change in the B_m_a_x of "3H-AMPA, though there was a two-fold increase in K_D, and little or no change in the immunoreactivity in GluR1, 2, 2/3 or 4. These results suggest that it is the regulation of receptors, their subunit composition and/or ...

Water consumption during solid state sodium borohydride hydrolysis

Energy Technology Data Exchange (ETDEWEB)

In this paper nickel acetate catalyzed sodium borohydride cartridges have been prepared and hydrolyzed with water for hydrogen production. Two technological solutions have been tested to increase the overall hydrogen yield, namely a porous water diffuser and a hydrophobic membrane. The first was used to improve water diffusion inside the hydride while the second to confine water inside the cartridge. The generated hydrogen flow showed a very reproducible behavior. Hydrogen promptly evolved just after water was pumped into the cartridge. After some initial peaks, a constant hydrogen flow has been recorded for the whole reaction time. The constant flow was related to the presence of the porous diffuser. The use of a hydrophobic membrane to confine the water inside the cartridge allowed to increase the overall hydrogen yield: about 6 water molecules per mol of hydride were required to complete the reaction. The reaction product was identified by XRD as Na{sub ...

Theoretical approach to initial growth kinetics of GaN on GaN(001)

British Library Electronic Table of Contents (United Kingdom)

We carried out theoretical analyses based on ab initio calculations incorporates in which free energy of the vapor phase is incorporated in order to determine the initial growth kinetics of c-GaN on GaN(001)-(4x1). The feasibility of the theoretical approach had been confirmed by calculations of Ga adsorption-desorption transition temperature and transition beam equivalent pressures on the GaAs(001)-(4x2)b2 surface in our previous work [Y. Kangawa, T. Ito, A. Taguchi, K. Shiraishi, T. Ohachi, Surf. Sci. 493 (2001) 178]. The results of calculations suggest that no Ga adsorption occurs on the initial surface under typical growth conditions but that a Ga adsorption site appears after N adsorption on GaN(001)-(4x1). That is, in the initial growth stage of c-GaN on GaN(001)-(4x1), a N-adsorbed ...

Structure of Ce-doped Bi/sub 2/(MoO/sub 4/)/sub 3/ as determined by neutron profile refinement

Energy Technology Data Exchange (ETDEWEB)

The structure of Bi/sub 1.8/Ce/sub 0.2/(MoO/sub 4/)/sub 3/ has been refined with powder neutron diffraction data by the Rietveld method. The structure can be derived by severely distorting the scheelite structure (AMO/sub 4/) and is perhaps better written A/sub (2/3)/phi/sub (1/3)/MO/sub 4/, where phi = cation vacancy. Of the two bismuth atom sites, cerium preferentially occupies the more symmetric of the two (Bi(2) in the structure) with some cerium found in the scheelite subcell vacancies also. This site preference is understood by examining the symmetries of the two Bi sites. Crystal data: monoclinic, space group P2/sub 1//c, Z = 4, a = 7.697(2), b = 11.535(3), c = 11.944(3), ..beta.. = 115.19.

Recommendations to the NRC for review criteria for alternative methods of low-level radioactive waste disposal: Task 2b: Earth-mounded concrete bunkers

International Nuclear Information System (INIS)

The US Army Engineers Waterways Experiment Station (WES) and US Army Engineer Division, Huntsville (HNDED) have developed general design criteria and specific design review criteria for the earth-mounded concrete bunker (EMCB) alternative method of low-level radioactive waste (LLW) disposal. An EMCB is generally described as a reinforced concrete vault placed below grade, underneath a tumulus, surrounded by filter-blanket and drainage zones. The tumulus is covered over with a low permeability cover layer and top soil with vegetation. Eight major review criteria categories have been developed ranging from the loads imposed on the EMCB structure through material quality and durability considerations. Specific design review criteria have been developed in detail for each of the eight major categories. 63 refs., 13 figs., 2 tabs.

Neutron shielding and constructional characteristics of a new type concrete and from borated clinker

International Nuclear Information System (INIS)

A boron containing cement, which can be used as nuclear shielding material, is produced at pilot plant scale applying two different methods. In the first method, the raw mixture of a normal portland cement is mixed with pre-calcined colemanite, a calcium borate mineral, and clinkerized in a rotary kiln (borated-clinker). In the second method, the colemanite is mixed with an admixture, which contains mainly limestone and marl, and burnt in the rotary kiln to obtain a borated-lime composite. The borated-lime composite is then added to the normal portland cement clinker up to 2% B_2O_3 content for shielding purpose. The results have shown that the borated-clinker contained untolerable amount of free lime resulting in a decrease in compressive strength. The addition of the borated-lime composite to the normal portland cement clinker up to 1% B_2O_3 content did not alter the setting time and the volume expansion properties. The reduction in the ...

K"A"T"P channels in mesenchymal stromal stem cells: Strong up-regulation of Kir6.2 subunits upon osteogenic differentiation

British Library Electronic Table of Contents (United Kingdom)

The promising use of mesenchymal stromal cells (MSC) in regenerative technologies accounts for necessity of detailed study of their physiology. Proliferation and differentiation of multipotent cells often involve changes in their metabolic state. In the present study, we analyzed the expression of ATP-sensitive potassium (K"A"T"P) channels in MSC and upon in vitro differentiation. K"A"T"P channels are present in many cells and regulate a variety of cellular functions by coupling cell metabolism with membrane potential. Kir6.1, Kir6.2 and SUR2A were expressed in undifferentiated MSC, whereas SUR2B and SUR1 were not detected on cDNA and protein level. Upon adipogenic differentiation Kir6.1 and SUR2A showed a significant reduction of the amount of mRNA by 84% and 95%, respectively, whereas Ki...

Interactions between organic anions on multiple transporters in Caco-2 cells.

Science.gov (United States)

In drug development, Caco-2 cells are often employed to study the influence of membrane transporters on drug permeability. The aim of the current study was to characterize permeability and kinetic parameters of selected organic anionic compounds in Caco-2 cells, and to investigate whether the Caco-2 cell line may be used as an overall model to predict interactions on multiple membrane transporters in the intestine. Taurocholic acid (TCA) and estrone-3-sulfate (E(1) S) were used as model substrates. Possible inhibitors studied were TCA, E(1) S, taurolithocholic acid, fluvastatin, and glipizide. The effects of these compounds on initial uptake, apparent permeability, and intracellular end-point accumulations of the probe substrates were studied. Both interactions on apical and basolateral influx transporters were observed. These interactions were proposed to be mediated mainly by the apical sodium-dependent bile acid transporter and the organic solute transporter ?/?, and to less extent ...

Histamine Molecule and Dianion Oxalate are Efficient Blocks for Building 2D Supramolecular Networks

British Library Electronic Table of Contents (United Kingdom)

One salt and two Cu(II) complexes (H2hsm)(ox), 1, [Cu(hsm)(ox)], 2, and [Cu(hsm)(ox)H2O], 3, have been synthesized and X-ray characterized (hsm is histamine and ox2? is the oxalate dianion). Starting from the prochiral tetracoordinated complex 2, pentacoordinated complex 3 crystallizes as a racemic mixture of the enantiomeric ? and ? isomers, in space group P21/c. In all cases, the side chain of the hsm group is gauche, allowing the formation of strong hydrogen bonds in the salt 1, and to chelate the metal center in complexes 2 and 3. The combination hsm/ox seems to favor the formation of 2D supramolecular structures (planes or wavy planes), through efficient networks of N?H???O hydrogen bonds. High resolution sequence stratigraphy, sedimentology, petrology and reservoir potentials of the Glauconitic member in Westerose and adjacent fields, Alberta, Canada

Energy Technology Data Exchange (ETDEWEB)

This thesis studied the petrology and sequence stratigraphy of the Hoadley barrier, a Glauconitic member in the Westerose field in Alberta. The unit contains gas reserves in an extensive northeast-southwest trending marine complex. A high resolution sequence stratigraphic model was used to understand the facies development of the unit and the diagenetic pathway of the strata. Six high frequency sequences were identified within the Glauconitic member. Each are separated from one another by surfaces of incision or subaerial exposure which sometimes coincide with a flooding surface and erosion. This thesis provided a petrographic characterization and depositional history of each of the 6 sequences. Reservoir quality is developed within the relatively quartz-rich foreshore facies of the G2B sandstones. The G3 chert litharenite to sublitharenite sandstones are excellent gas producers because of their coarser grain size and secondary porosity. The ...

Design of modular coils for a quasi-axisymmetric stellarator with a flexible control of the magnetic field configuration

Energy Technology Data Exchange (ETDEWEB)

A design of the modular coil system for CHS-qa has been made for the plasma configuration '2b32' with the aspect ratio 3.2. The magnetic field strength and the major radius are 1.5 T and 1.5 m, respectively. The normal component of magnetic field produced by the modular coils is minimized on the plasma boundary to obtain the optimum coil design. We put engineering constraint on the distance between adjacent modular coils and the radius of coil curvature. The dependence of the residual normal component of the field on these conditions is examined, and the realistic values for them are selected. Additional coils to control various properties of the magnetic field configuration (the rotational transform, the magnetic well depth, etc.) have been designed and a flexibility of the magnetic field configuration is realized. For the case that the rotational transform crosses the low-order rational value resulting in magnetic islands, ...

Controllable synthesis of graphene sheets with different numbers of layers and effect of the number of graphene layers on the specific capacity of anode material in lithium-ion batteries

International Nuclear Information System (INIS)

High quality graphene sheets are synthesized through efficient oxidation process followed by rapid thermal expansion and reduction by H2. The number of graphene layers is controlled by tuning the oxidation degree of GOs. The higher the oxidation degree of GOs is getting, the fewer the numbers of graphene layers can be obtained. The material is characterized by elemental analysis, thermo-gravimetric analysis, scanning electron microscopy, atomic force microscopy, transmission electron microscopy and Fourier transform infrared spectroscopies. The obtained graphene sheets with single, triple and quintuplicate layers as anode materials exhibit a high reversible capacity of 1175, 1007, and 842 mA h g-1, respectively, which show that the graphene sheets with fewer layers have higher reversible capacity. -- Graphical abstract: The typical TEM images of the graphene sheets derived from GO3(a), GO2(b) and GO1(c). Cloning and expression of a human kidney cDNA for an /alpha//sub 2/-adrenergic receptor subtype

Energy Technology Data Exchange (ETDEWEB)

An /alpha//sub 2/-adrenergic receptor subtype has been cloned from a human kidney cDNA library using the gene for the human platelet /alpha//sub 2/-adrenergic receptor as a probe. The deduced amino acid sequence resembles the human platelet /alpha//sub 2/-adrenergic receptor and is consistent with the structure of other members of he family of guanine nucleotide-binding protein-coupled receptors. The cDNA was expressed in a mammalian cell line (COS-7), and the /alpha//sub 2/-adrenergic ligand (/sup 3/H)rauwolscine was bound. Competition curve analysis with a variety of adrenergic ligands suggests that this cDNA clone represents the /alpha//sub 2/B-adrenergic receptor. The gene for this receptor is on human chromosome 4, whereas the gene for the human platelet /alpha//sub 2/-adrenergic receptor (/alpha//sub 2/A) lies on chromosome 10. This ability to express the receptor in mammalian cells, free of other adrenergic receptor subtypes, should help ...

Affinity purification of sequence-specific DNA binding proteins.

Science.gov (United States)

We describe a method for affinity purification of sequence-specific DNA binding proteins that is fast and effective. Complementary chemically synthesized oligodeoxynucleotides that contain a recognition site for a sequence-specific DNA binding protein are annealed and ligated to give oligomers. This DNA is then covalently coupled to Sepharose CL-2B with cyanogen bromide to yield the affinity resin. A partially purified protein fraction is combined with competitor DNA and subsequently passed through the DNA-Sepharose resin. The desired sequence-specific DNA binding protein is purified because it preferentially binds to the recognition sites in the affinity resin rather than to the nonspecific competitor DNA in solution. For example, a protein fraction that is enriched for transcription factor Sp1 can be further purified 500- to 1000-fold by two sequential affinity chromatography steps to give Sp1 of an estimated 90% homogeneity with 30% yield. ...

A study on heat-transfer enhancement by a square-rod array in an impinging jet system

Energy Technology Data Exchange (ETDEWEB)

An impinging jet is a widely used technique for realizing high heat-transfer rates between a fluid and a surface. However, the area of enhanced heat transfer is limited to the neighborhood of the stagnation point. In this study, heat transfer is augmented remote from the stagnation point in an impinging plane jet system by a rod array located near the wall. Each square rod in the array was positioned normal to the flow direction and parallel to the flat plate surface. The distance between the nozzle and the flat plate (H) and the spacing between the rods and the flat plate surface (C) were changed to find the optimum values. The largest heat-transfer augmentation was obtained for C = 1 mm, H/B = 10, where the jet nozzle width is B. In this case, the heat-transfer coefficient averaged over an area 2B from the stagnation point is about 1.6 times greater compared to that without a rod array.

Transient enhanced diffusion from decaborane molecular ion implantation

International Nuclear Information System (INIS)

Transient enhanced diffusion (TED) from implantation of 5keVB_1_0H_1_4 and 0.5 keV B ions has been quantified and compared for nominal boron doses of 10"1"4 and 10"1"5cm"-"2. Boron diffusivity during annealing was extracted from secondary ion mass spectroscopy depth profiles of diffused marker layers in boron doping-superlattices and the actual implanted B dose was independently measured by nuclear reaction analysis. Comparable enhancements were observed from both ions. Transmission electron microscopy analysis revealed that both boron- and decaborane-implanted samples were amorphized at a nominal 10"1"5cm"-"2B dose. A comparison with data from low energy Si implants revealed a similar dependence of diffusivity enhancement on implant dose. These findings are consistent with the understanding that TED is caused by the interstitial supersaturation resulting from a number of excess interstitials approximately equal to the number of implanted atoms ...

Temperature evolution of the internal magnetic hyperfine field of Metglas: amorphous and crystallized phases

International Nuclear Information System (INIS)

We present differential scanning calorimetry (DSC), X-ray diffractometry, in situ Moessbauer spectroscopy (MS), and transmission electron microscopy (TEM) studies in Metglas ribbons subjected to different heat treatments. The temperature evolution of the hyperfine field H_h_f(T) and the Curie temperature (T_c) of the amorphous phase are determined. The magnetic field originally present in the amorphous phase has a 'normal' behavior, in the sense that it can be described by the Weiss molecular field theory. The total angular momentum of the iron atoms turns out to be 5/2 and this implies Fe"3"+ in which the electronic spins are uncoupled. When the samples are maintained near T_c (#approx#673 K), three new magnetic phases are detected in the Moessbauer spectra, indicating an onset of a crystallization process well below the first crystallization temperature (T_X_1), as determined by DSC (#approx#820 K). The magnetic behavior of these phases is also described by universal Brillouin curves ...

Physics of locked modes in ITER: Error field limits, rotation for obviation, and measurement of field errors

Energy Technology Data Exchange (ETDEWEB)

The existing theoretical and experimental basis for predicting the levels of resonant static error field at different components m,n that stop plasma rotation and produce a locked mode is reviewed. For ITER ohmic discharges, the slow rotation of the very large plasma is predicted to incur a locked mode (and subsequent disastrous large magnetic islands) at a simultaneous weighted error field ({Sigma}{sub 1}{sup 3}w{sub m1}B{sup 2}{sub rm1}){sup {1/2}}/B{sub T} {ge} 1.9 x 10{sup -5}. Here the weights w{sub m1} are empirically determined from measurements on DIII-D to be w{sub 11} = 0. 2, w{sub 21} = 1.0, and w{sub 31} = 0. 8 and point out the relative importance of different error field components. This could be greatly obviated by application of counter injected neutral beams (which adds fluid flow to the natural ohmic electron drift). The addition of 5 MW of 1 MeV beams at 45{degrees} injection would increase the error field limit by a factor ...

A study by the U.S. Air Force School of Aerospace Medicine, Brooks AFB, Texas, has compared the shelter processing transfer of chemical warfare agent simulant vapor for subjects wearing unwashed and laundered ground-crew chemical defense overgarments. Twice laundered and four times laundered protective garments were included in the assessment. Test subjects, wearing the unwashed and laundered protective garments were initially sprayed with liquid simulant (methyl salicylate) to a target density of 5 g m-2. They were then sequentially processed into and through the USAFSAM Collective Protection Shelter (SCPS-2B) test facility. Immediately upon entry to the Toxic-Free Area, the subjects were confined in individual off gassing booths for 2 h while offgassed simulant vapor concentration in the booths was recorded. The resulting mean maximum vapor concentrations recorded in the booths for subjects who had worn unwashed and laundered overgarments ...

Genetic analysis of carbon isotope discrimination and its relation to yield in a wheat doubled haploid population.

Science.gov (United States)

Carbon isotope discrimination (?(13) C) is considered a useful indicator for indirect selection of grain yield (GY) in cereals. Therefore, it is important to evaluate the genetic variation in ?(13) C and its relationship with GY. A doubled haploid (DH) population derived from a cross of two common wheat varieties, Hanxuan 10 (H10) and Lumai 14 (L14), was phenotyped for ?(13) C in the flag leaf, GY and yield associated traits in two trials contrasted by water availability, specifically, rain-fed and irrigated. Quantitative trait loci (QTLs) were identified by single locus and two locus QTL analyses. QTLs for ?(13) C were located on chromosomes 1A, 2B, 3B, 5A, 7A and 7B, and QTLs for other traits on all chromosomes except 1A, 4D, 5A, 5B and 6D. The population selected for high ?(13) C had an increased frequency of QTL for high ?(13) C, GY and number of spikes per plant (NSP) when grown under rain-fed conditions and only for high ?(13) C and NSP ...

We perform two-dimensional bulge-disk-bar decomposition on H-band images of 143 bright, high-mass (M _* #>=# 1.0 x 10"1"0 M _s_u_n) low-to-moderately inclined (i < 70"0) spirals. Our results are as follows. (1) Our H-band bar fraction (#approx#58%) is consistent with that from ellipse fits. (2) 70% of the stellar mass is in disks, 10% in bars, and 20% in bulges. (3) A large fraction (#approx#69%) of bright spirals have B/T#<=# 0.2, and #approx#76% have low n #<=# 2 bulges. These bulges exist in barred and unbarred galaxies across a wide range of Hubble types. (4) About 65% (68%) of bright spirals with n #<=# 2 (B/T #<=# 0.2) bulges host bars, suggesting a possible link between bars and bulges. Earlier polycrystal films with anomalous photo voltage properties were received only on the clean and flat surface with high isolation property (R>10"1"4 #OMEGA#). We worked out the technology of receiving APhV films CdTe on the surface of the photosensitive film CdS. The main condition under which this effect may be observed is that the lining must be slant concerning to the direction of the motion of evaporated substance's molecular bunch during the growing of the film, i.e. received film must have a dendritic structure. That is why photosensitive film CdS was received with thermo vacuum slanting ...

Crystal and electronic structures, luminescence properties of Eu2+-doped Si6-zAlzOzN8-z and MySi6-zAlz-yOz+yN8-z-y (M=2Li, Mg, Ca, Sr, Ba)

International Nuclear Information System (INIS)

The crystal structure, electronic structure, and photoluminescence properties of EuxSi6-zAlz-xOz+xN8-z-x (x=0-0.1, 0xMySi6-zAlz-x-yOz+x+yN8-z-x-y (M=2Li, Mg, Ca, Sr, Ba) have been studied. Single-phase EuxSi6-zAlz-xOz+xN8-z-x can be obtained in very narrow ranges of x?0.06 (z=0.15) and z2+ ions can be incorporated into nitrogen-rich Si6-zAlzOzN8-z. The Eu2+ ion is found to occupy the 2b site in a hexagonal unit cell (P63/m) and directly connected by six adjacent nitrogen/oxygen atoms ranging 2.4850-2.5089 A. The calculated host band gaps by the relativistic DV-X? method are about 5.55 and 5.45 eV (without Eu2+ 4f5d levels) for x=0 and 0.013 in EuxSi6-zAlz-xOz+xN8-z-x (z=0.15), in which the top of the 5d orbitals overlap with the Si-3s3p and N-2p orbitals within the bottom of the conduction band of the host. EuxSi6-zAlz-xOz+xN8-z-x shows a strong green emission with a broad Eu2+ band centered at about 530 nm under UV to near-UV excitation range. ...
